The probit 9 standard for quarantine treatment efficacy (99.9968% mortality) was originally recommended for tropical fruits heavily infested with fruit flies and it centers on high mortality to achieve quarantine security. This standard may be too stringent for quarantine pests in commodities that are rarely infested or are poor hosts. The alternative treatment efficacy approach measures risk as the probability of a mating pair, gravid female, or parthenogenic individual surviving in a shipment. This will be a function of many factors including infestation rate and shipment volume. Applying the risk-based alternative treatment efficacy approach to pests on rarely infested or poor hosts will lower the number of required test insects needed for developing quarantine treatments; hence data for a quarantine treatment could be generated by testing 10,000 or fewer insects with no survivors, compared with 90,000–100,000 insects to demonstrate the traditional probit 9 efficacy. Several commodity/quarantine pest systems where this approach could be applied are discussed. This approach would save time and resources, and help farmers export their crop on a more-timely basis.

Cultural control methods have been central in the southwestern United States for reducing pink bollworm, Pectinophora gossypiella (Saunders), damage to cotton. Nevertheless, it is not clear at present how such methods could be integrated within the novel pest management framework allowed by introduction of cotton producing a toxin from Bacillus thuringiensis (Bt) for pink bollworm control. Using statewide pheromone trapping and climatic data in conjunction with deterministic simulation models, we investigated whether manipulation of cotton planting date and use of other cultural control methods could represent valuable tactics for control of the pink bollworm in Arizona. Accumulation of heat units from one January accurately predicted the rate of pink bollworm emergence from diapause in 15 cotton-producing regions. Significant variation in rate of emergence from diapause was present among regions, with earlier emergence at higher altitudes. Most adults emerge from diapause too early to reproduce successfully on cotton, a phenomenon known as suicidal emergence. A method for prediction of the fraction of suicidal emergence resulting from adoption of a given cotton planting date is presented. Results from simulation models suggest that manipulation of planting date and implementation of other control cultural methods reduce the rate of application of insecticides and delay the evolution of resistance to Bt cotton in the pink bollworm.

Female mites of the genus Varroa reproduce on the immature stages of Apis cerana F. and A. mellifera L. Mites are found more often in drone brood than worker brood, and while evolutionary explanations for this bias are well supported, the proximate mechanisms are not known. In one experiment, we verified that the proportion of hosts with one or more mites (MPV, mite prevalence value) was significantly greater for drones (0.763 ± 0.043) (lsmean ± SE) than for workers (0.253 ± 0.043) in populations of mites and bees in the United States. Similar results were found for the average number of mites per host. In a second experiment, using a cross-fostering technique in which worker and drone larvae were reared in both worker and drone cells, we found that cell type, larval sex, colony and all interactions affected the level of mites on a host. Mite prevalence values were greatest in drone larvae reared in drone cells (0.907 ± 0.025), followed by drone larvae reared in worker cells (0.751 ± 0.025), worker larvae reared in worker cells (0.499 ± 0.025), and worker larvae reared in drone cells (0.383 ± 0.025). Similar results were found for the average number of mites per host. Our data show that mite levels are affected by environmental factors (cell type), by factors intrinsic to the host (sex), and by interactions between these factors. In addition, colony-to-colony variation is important to the expression of intrinsic and environmental factors.

Homalodisca coagulata (Say) is a recent introduction to California. It is known to spread a strain of the bacterium Xylella fastidiosa Wells, Raju, Hung, Weisberg, Mandelco-Paul & Brenner that induces oleander leaf scorch disease in oleander, Nerium oleander L. Oleander leaf scorch is lethal to oleander and threatens to decimate one of the most important landscape shrubs in California. Towards developing a management strategy for H. coagulata-spread oleander leaf scorch, we documented the affects of selected insecticides on H. coagulata mortality, feeding behavior, and disease transmission in a greenhouse study. Oleanders treated with fenpropathrin, fenpropathrin acephate, and imidacloprid caused significant mortality to caged H. coagulata within 4 h of exposure. Within 24 h, these pesticides caused nearly 100% mortality 3 wk after treatment. In other experiments, acetamiprid and fenpropathrin treatments reduced time spent feeding and total time on plants. H. coagulata on fenpropathrin-, acetamiprid-, and imidacloprid-treated oleander died in less than 13 min on average. Oleander leaf scorch transmission by H. coagulata was blocked by applications of foliar-applied acetamiprid, and soil-applied imidacloprid and thiamethoxam.

Nucleopolyhedroviruses originally isolated from Anagrapha falcifera (Kirby) and Autographa californica (Speyer) were formulated with various ingredients using a spray dry method and tested for residual field activity in Illinois and Mississippi. In Mississippi, field tests were conducted on cotton in 1997, whereas in Illinois tests were conducted on cabbage in 1997 and 1998. Within 24 h, significant differences were observed among formulations in all tests. Unformulated virus had significantly less insecticidal activity than formulated virus and formulations containing lignin retained activity significantly longer than other formulations. Relatively small amounts of Blankophor BBH, when encapsulated within the formulation, did not greatly enhance (>10×) insecticidal activity based on LC₅₀ determinations nor prolong insecticidal activity based on field evaluations. In most tests, >50% activity remained in formulations containing lignin, whereas unformulated virus retained <50% activity within 24 h after application.

Studies were performed in the laboratory, greenhouse and field to assess the potential of Agrotis ipsilon multicapsid nucleopolyhedrovirus (AgipMNPV) and a viral enhancing agent, M2R, for suppression of Agrotis ipsilon (Hufnagel). In laboratory droplet feeding bioassays, AgipMNPV was shown to be highly active against third-instar A. ipsilon. The optical brightener M2R significantly reduced LD₅₀ estimates by ≈160-fold, but had no direct effect on survival time estimates. In greenhouse trials, spray and bait formulations of AgipMNPV significantly reduced feeding damage to corn seedlings caused by third-instar A. ipsilon. In two sets of replicated field trials, bait formulations of AgipMNPV significantly reduced feeding damage to corn seedlings by third-instar A. ipsilon. However, there were no beneficial effects attributable to the inclusion of M2R in AgipMNPV formulations under greenhouse or field conditions. It seems likely that in an appropriately designed pest management program AgipMNPV could be used to suppress field populations of early and mid-instar A. ipsilon.

The effects of a transgenic Bacillus thuringiensis (Bt)-cotton cultivar (DPL 32) on three instars of the soybean looper, Pseudoplusia includens (Walker), were determined in laboratory studies. First, third, and fifth instars were fed field collected Bt-cotton leaves for 1, 2, four and 7 d or until pupation, and then transferred to artificial diet. Mortality during the larval stage increased linearly in response to an increase in the length of feeding time on Bt-cotton by first and third instars. The maximum mortality of about two out of three larvae occurred for first instars fed on Bt-cotton until pupation. For the fifth instar, there was no significant response to feeding time; however, most of these larvae reached pupation before 4 d of feeding on Bt-cotton. The length of the larval developmental period also increased linearly with an increase in feeding time on Bt-cotton in first and third instars; again, there was no significant response in the fifth instars. For both mortality and larval developmental time, the linear trend lines for the first and third instars were quite similar. Pupal weight declined linearly in the first and fifth instars in response to feeding time on Bt-cotton. Although pupal weight also declined for third instars, the response was not linear. The effect of Bt-cotton appears not to extend past pupation in that there were no significant responses in mortality and developmental time of pupae during the pupal stage. These data indicate that larvae surviving Bt-cotton are adversely affected in several ways, which should be considered in evaluating Bt-cotton suppression of soybean looper infestations.

The fruit fly Dacus ciliatus Loew is a pest of the fruits of many cucurbit species. We studied the effect of organophosphate and pyrethroid compounds on the adult flies by using surface contact and oral administration. In contrast to other fruit flies, we found that organophosphates were ineffective against D. ciliatus. This was supported by the insignificant decrease of head acetylcholinesterase activity. All tested pyrethroids showed satisfactory killing ability, rapid and massive knockdown effect, and prevention of oviposition. Piperonyl butoxide considerably increased the toxicity of pyrethroids, which can be explained by oxidase detoxification of these compounds in D. ciliatus. It can be concluded that pyrethroids have high potential for controlling D. ciliatus.

Incidence of insect damage, and association of insect damage with mycotoxigenic corn ear molds and mycotoxins was examined in commercial fields of Bt and non-Bt hybrids of different backgrounds in Illinois in 1998 and 1999. Nearly 50% Helicoverpa zea (Boddie) infestation sometimes occurred in Bt hybrids that express high levels of the protein in silks and kernels. Damage by European corn borer, Ostrinia nubilalis Hübner, was uncommon, even in non-Bt ears. Levels of total fumonisins were generally less (15- to 1.8-fold) in Bt versus non-Bt hybrids at the same site, with some significant differences. There were several instances where there were no significant differences in fumonisin levels between low/no Bt kernel hybrids and Bt hybrids that produced high levels of the protein in the kernel and silk tissue. However, significant correlations were often noted between numbers of insect-damaged kernels and total fumonisin levels, especially in 1998, suggesting in these cases that reducing insect damage was still reducing fumonisin levels. There was variability between the correlation coefficient for numbers of insect damaged kernels and fumonisin levels at different sites for the same year, different hybrids at the same site, and the same hybrid for different years. Although reductions in fumonisins in Bt hybrids were more limited than reported in the past, planting the Bt hybrids still appears to be a useful method for indirectly reducing mycotoxins in corn ears.

Pesticides were evaluated for their effect on two parasitoid species, Colpoclypeus florus and Trichogramma platneri, that are potential biological control agents of leafrollers in apple orchards. Organophosphate and carbamate insecticides were highly toxic to both parasitoids in topical applications, but foliar residues of some products were nontoxic after 7 d. At reduced rates, topically applied pyrethroids were low in toxicity to C. florus were highly toxic to T. platneri, and foliar residues were nontoxic after about 7 d. Imidacloprid and abamectin were highly toxic when applied topically to both parasitoids but were not toxic as 1-d-old residues. Insect growth regulators did not cause mortality either as topical applications or residues; however, diflubenzuron caused severe sublethal effects, completely blocking the production of C. florus offspring. Biorational pesticides, such as soap, oil, and B. thuringiensis products, caused no toxicity to C. florus but had a direct impact on T. platneri as topical applications through physical immobilization. The potential to integrate different pesticides with biological control of leafrollers and the need for a step-wise approach to evaluate the impact of pesticides against natural enemies is discussed.

Helicopter applications using sabadilla (27–57 g [AI]/ha and 11.1–23.7 kg/ha sugar bait) in different volumes of water were made on avocado against Scirtothrips perseae Nakahara in Ventura County, CA. On small (2.8 m tall) trees, volumes of 374 and 795 liter/ha resulted in 3–18% and 19–43% spray coverage, respectively, on water-sensitive papers. On large (6.9–8.4 m tall) trees, these volumes resulted in only 1–3% and 6–11% coverage, respectively. On small trees, sabadilla in 374 liter/ha significantly reduced numbers of larvae 3 and 6 d after treatment (DAT). In contrast, on large trees, 374 liter/ha did not reduce larval numbers and was less effective than 561–1,964 liter/ha from 3–11 DAT. However, volumes of 1,543 and 1,964 liter/ha did not increase efficacy compared with 1,169 liter/ha. The results indicate that large trees require sabadilla in higher spray volumes for control of S. perseae, making applications on large trees more costly. The extra costs may not result in larval control beyond 11 d, making it especially critical to properly time applications on large trees.

Helicopter applications using abamectin in different spray volumes were made against Scirtothrips perseae Nakahara in Ventura County, CA. On small (2.2 m tall) trees, spray coverage on water-sensitive papers was 24–48% and 43–97% for 468 and 935 liter/ha volume treatments, respectively. On large (6.2–8.1 m tall) trees, spray coverage was lower and quite variable, from 1 to 28% and 10 to 70% for 468 and 935 liter/ha treatments, respectively. On small trees, 468, 701, and 935 liter/ha with a high abamectin rate (26 g [AI]/ha) were equally effective against larvae from 13 to 27 d after treatment (DAT). On medium (4.2 m tall) trees, 468 and 935 liter/ha with the high rate were equally effective from 23 to 113 DAT. On large (6.5–8.1 m tall) trees, 468 and 935 liter/ha with a low abamectin rate (13 g [AI]/ha) were ineffective in three tests. In a fourth large (6.8 m tall) tree test, 468 and 935 liter/ha with the high rate were effective at 3 and 37 DAT. In a fifth large (6.2 m tall) tree test, 468-1, 403 liter/ha with the high and 935 liter/ha with the low rate were equally effective 2-22 DAT. After all effective treatments, thrips numbers were lower than in controls for 1–3 mo. However, stable and highest reductions in populations were sometimes delayed until 20–23 DAT even when coverage was high. The variability in spray coverage on the lower levels of large trees and the delayed effect may explain inconsistencies in the reporting of or in actual aerial application results.

Pot experiments were used to investigate the effect of root-feeding larvae of the soldier fly Inopus rubriceps (Macquart) on shoot production from sugarcane planting pieces (setts) and on growth and ratooning of sugarcane plants. Shoot elongation was inhibited while setts were exposed to larvae, and it resumed when larvae were removed. Infested setts produced a greater weight of roots than uninfested setts. Similar symptoms were induced by mechanical root pruning, suggesting that the effect of soldier fly larvae on setts may be a redirection of growth from the shoot to roots due to root damage. Larvae had a greater effect on shoot production at lower temperature, particularly in cultivar ‘Q151’, which had a higher temperature threshold than ‘CP44–101’. Temperature and cultivar may influence the harmful effect of soldier fly larvae on sett germination by changing the differential rates of plant growth and larval feeding. When growing plants were exposed to larvae, the infested plants were slightly smaller at harvest and subsequently produced many fewer ratoon shoots from underground buds than uninfested plants. Shoot elongation from buds was also inhibited in setts cut from the above-ground stalks of infested plants. Analysis of nutrient levels in plants did not indicate the mechanism for ratooning inhibition, because levels of the 10 elements analyzed were at least as high or higher in infested plants. Infestation was associated with an increased level of sucrose and a reduced level of fructose in stalks. The inhibitory effect of larval feeding on ratooning was not reversed when larvae were removed from pots 10 wk before harvest. However, new stubble produced from infested plants then ratooned normally after a second harvest, provided the new roots were not attacked. The symptoms of larval feeding in growing plants are unexplained, but may be caused by the prolonged withdrawal of sap from roots or the injection of some inhibitory substance by larvae.

Field sampling methods and economic thresholds were developed to provide management recommendations for Helicoverpa armigera (Hübner) on processing tomatoes, based on a commercially acceptable damage level of 5% fruit damage. Population estimates from destructive sampling and a rapid 1-min plant scouting method were related to fruit damage, and a nominal economic threshold of one larva per plant was derived. The economic threshold was confirmed in a designed trial where it resulted in acceptable levels of fruit damage. The 1-min scouting method and economic threshold was validated in 17 commercial crops in the Gisborne, Poverty Bay region of New Zealand. Scouting in these fields was based on 10 plants in each of four quadrants proportionally representing the topography of each field. In unsprayed areas, egg and larval populations were usually below the economic threshold in early-planted crops but often exceeded thresholds in late-planted crops. In commercial demonstration trials where standard calendar spraying practice was compared with no spraying, calendar-based applications maintained fruit damage below 3.4%, but insecticide was applied unnecessarily to more than half the crops. Larval populations were a significant predictor of damage in these commercial crops. In 12 implementation trials, where spraying recommendations were based on the 1-min scouting threshold of one larva per plant, the worst fruit damage observed was 2.3%. The definition of an economic threshold, scouting methods, and establishment of parasitoids have reduced spray applications and contributed to the implementation of an integrated pest management program for processing tomatoes in New Zealand.

Root and lower stem insects cause significant damage to conifers, vector phytopathogenic fungi, and can predispose trees to bark beetle attacks. The development of effective sampling techniques is an important component in managing these cryptic insects. We tested the effects of trap type and stereochemistry of α-pinene, in combination with ethanol, on catches of the root colonizing weevils (Coleoptera: Curculionidae) Hylobius spp. [mostly Hylobius pales (Herbst)], and Pachylobius picivorus (Germar), the root colonizing bark beetle (Coleoptera: Scolytidae) Hylastes porculus Erickson, and the lower stem colonizing bark beetle Dendroctonus valens (LeConte). We tested for inter-regional differences by conducting similar field assays in the northern (Wisconsin) and southern (Louisiana) United States. The more effective trap type varied with region. Root weevils were caught primarily in pitfall traps in Wisconsin, whereas they were caught mostly in lower stem flight traps in Louisiana. In Wisconsin, root colonizing bark beetles were also caught primarily in pitfall traps, but lower stem colonizing bark beetles were caught primarily in lower stem flight traps. The root feeding weevils preferred (−) over ( )-α-pinene in both regions. Some exceptions relating to trap type or gender occurred in southern populations. The two root and lower stem colonizing bark beetles in Wisconsin showed no preference between ( ) and (−)-α-pinene in combination with ethanol. No bark beetles were caught in the south. Our results suggest that modifying trap type and enantiomeric ratios of monoterpenes for different insect groups and in different regions can improve sampling efficiency for these important pests.

Currently, there is an elevated interest in reducing feeding damage caused by the Nantucket pine tip moth, Rhyacionia frustrana (Comstock), a common regeneration pest of loblolly pine, Pinus taeda L. The toxicity of several insecticides was tested in a laboratory against four common R. frustrana parasitoids. There were no differences in parasitoid mortality between the control and indoxacarb treatments. However, the pyrethroids, permethrin and lambda-cyhalothrin, caused significantly more mortality initially (up to 240 min exposure time) than other insecticides. Spinosad was less toxic than the pyrethroids initially, but the spinosad related mortality increased with time until it reached a level similar to the pyrethroids. For the most part, spinosad and the pyrethroids caused more mortality than the control and indoxacarb treatmtents within the 1-d sample period. These results may have important implications for decisions concerning which insecticides are best suited for reducing pest damage while conserving natural enemies in timber and agricultural systems. Large-scale field trials are required to further define the impacts of these insecticides on natural enemies.

Macrolepidopteran caterpillars collected in 1995 and 1996 in the Monongahela National forest, Pocahontas County, WV, and the George Washington National forest, Augusta County, VA, yielded 60 previously unreported tachinid host associations. Most associations were between native species, but the introduced polyphagous tachinid Compsilura concinnata (Meigen) produced eight new associations with native hosts. The tachinids collected were slightly broader in their host preferences than associated Braconidae and Ichneumonidae, illustrating their potential importance in regulating foliage-feeding macrolepidopteran populations in the region studied. The sample years reported herein are the pretreatment baseline portion of a long-term study on the effects of Lymantria dispar (L.) defoliation, efficacy of applied Bacillus thuringiensis variety kurstaki Berliner, and the natural occurrence of the L. dispar pathogen Entomophaga maimaiga Humber, Shimazu & Soper on nontarget organisms.

Host-mediated effect on the efficacy of Bacillus thuringiensis Berliner against larvae of the forest tent caterpillar, Malacosoma disstria Hübner, was investigated under controlled conditions. Host plants used in this study were quaking aspen, Populus tremuloides Michx., a preferred host, and sugar maple, Acer saccharum Marsh., a secondary host. Larvae were reared in the laboratory on leaves of these hosts, and upon reaching the third, fourth, and fifth instar, they were fed leaves treated with one of a range of concentrations of B. thuringiensis variety kurstaki HD-1 suspensions. Larvae were tested on the host on which they were feeding before the 4-d bioassays. The estimated LC₅₀s were 100-fold greater on quaking aspen than on sugar maple. Also, there was a decrease in efficacy over the whole ranges of concentrations with larval age on both hosts. LC₅₀s varied approximately two-fold between third and fifth instar. These results indicate that host-mediated effects on B. thuringiensis efficacy warrant more interest. In particular, they strongly indicate that the host plant modifies the interaction between B. thuringiensis and a target insect, and offer the opportunity to investigate the mechanism(s) that may be involved in the enhancement of B. thuringiensis toxicity.

Field-based studies and laboratory bioassays were conducted with apple maggot, Rhagoletis pomonella (Walsh), and blueberry maggot, Rhagoletis mendax Curran, flies to investigate the performance and duration of activity of insecticide-treated biodegradable and wooden spheres for control of Rhagoletis species. Four neonicotinoid insecticide treatments including imidacloprid, thiamethoxam, and thiocloprid at 2% (AI) were evaluated with biodegradable spheres. In 1999, significantly more apple maggot flies were found killed by imidacloprid-treated spheres compared with thiamethoxam-treated spheres during early and late season. In 2000, spheres treated with either of two formulations of imidacloprid killed significantly more apple maggot flies compared with thiamethoxam, thiocloprid, and untreated spheres. In blueberries, there were no significant differences between the numbers of blueberry maggot flies killed by both imidacloprid-treated or thiamethoxam-treated spheres in 1999. However, during the 2000 blueberry field season, both formulations of imidacloprid were significantly more effective in killing blueberry maggot flies compared with spheres treated with thiamethoxam, thiocloprid and untreated controls. Overall, spheres treated with thiocloprid were ineffective and did not kill significantly more apple maggot or blueberry maggot flies compared with the controls. Laboratory bioassays showed that the effectiveness of field-exposed spheres treated with imidacloprid at 4 and 8% (AI) and thiamethoxam at 4% (AI) in killing apple maggot flies was not significantly reduced over a 12-wk aging period. Additionally, wooden spheres aged outdoors for 12 wk with and without mold maintained residual activity in laboratory tests, whereas biodegradable spheres of equal aging, with and without mold lost their effectiveness in killing apple maggot flies. In other studies, we confirmed that the addition of an external feeding stimulant (sucrose) significantly increases the effectiveness of both biodegradable and wooden spheres treated with imidacloprid at 2% (AI).

Numerous field studies were conducted in commercial nurseries in Tennessee from 1996 through 1999 to evaluate chemical and biological treatments, application timing and rates, and method of application for control of early instars of Japanese beetle, Popillia japonica Newman. Insecticide treatments included bifenthrin, bendiocarb, chlorpyrifos, carbaryl, fipronil, halofenozide, imidacloprid, permethrin, tefluthrin, thiamethoxam, and trichlorfon. Biological treatments included entomopathogenic nematodes (Heterorhabditis bacteriophora HP88 or H. marelatus), Bacillus thuringiensis Berliner subspecies japonensis Buibui strain, and Beauveria bassiana (Balsamo) Vuillemin. All treatments were applied on the soil surface or injected into the soil around the base of each tree. Tree type and size varied among and within tests, however, the sampling unit (61-cm-diameter root ball) remained the same throughout all tests. The biological treatments provided poor-to-moderate control (0–75%) of Japanese beetle larvae. Imidacloprid was the most frequently evaluated insecticide and achieved 91–100, 87–100, 83–100, and 41–100% control with applications in May, June, July, and August, respectively. Halofenozide treatments were not significantly different from imidacloprid treatments with one exception. Halofenozide provided 60–87, 85–100, and 82–92 control with applications made in June, July, and August, respectively. Fipronil and thiamethoxam were evaluated to a lesser extent but both performed similarly to imidacloprid. Most other insecticide treatments were less successful in reducing numbers of Japanese beetle larvae and with few exceptions achieved <50% control.

The response of potato, Solanum tuberosum L., tuber yield to stem injury by European corn borer, Ostrinia nubilalis (Hübner), larvae was investigated in the Mid-Atlantic region of the United States for 3 yr. This response was described for ‘Superior’, ‘Atlantic’, and ‘Snowden’ potato, which are early-season, midseason, and late-season maturing cultivars, respectively. To model the yield/injury relationship, a range of corn borer injury levels was established in the field by augmenting the natural infestation with varying densities of laboratory-reared larvae. Linear and nonlinear regression analyses (plateau and second-order polynomial models) were used to describe the relationship between yield of U.S. No. 1 grade tubers and the percentage of stems injured by corn borer larvae. The maturity of the cultivar did not affect the response of potato yield to stem injury. In nine of 14 experiments, potato tolerated high levels of corn borer injury (55–90% of stems injured) without yield loss, suggesting that control of corn borer may not be necessary. Yet, in one of five Superior tests, in two of four Atlantic tests and in two of five Snowden tests, corn borer injury significantly reduced yield. Of the five data sets in which corn borer injury reduced yield, the plateau model fit two data sets and the quadratic model fit one data set. In two of the three cases, these models accounted for nearly identical amounts of total variation in yield as that accounted for by the linear model. The linear model fit four of the five data sets, but the R² values were low for three of the four tests (0.10, 0.18, and 0.31). The parameter or parameters that interact with corn borer injury to cause tuber yield reduction should be identified before economic injury levels and thresholds are developed.

Some recent high-load, low-density pheromone-release devices emit an ethanolic blend of pheromone directly onto crop foliage to control insect pests by mating disruption. This study characterized the phytotoxicity associated with deposition of some pheromonal compounds in concentrated drops on the foliage of trees bearing aerosol release devices. The relative toxicity of straight-chained alkanes, alcohols, aldehydes, and acetates with chain lengths varying from C-2 to≈C-20 was quantified in the laboratory by the severity of necrotic lesions. The order of severity for phytotoxicity caused by pheromonal compounds was alkanes ≪ acetates = aldehydes ≤ alcohols. Within compound classes tested, pheromones with chain lengths of 6–13 carbons were the most phytotoxic. Phytotoxicity was not detectable at dosages <1 mg administered in 10 μl of ethanol. Phytotoxicity of pheromones was highly correlated with presence of both a hydrophilic and lipophilic molecular domain. We postulate nonspecific membrane disruption as a likely mode of action for pheromonal phytotoxicity. Limited attempts to remediate this effect by changing carrier solvents or adding surfactants, spreaders, or nonvolatile diluents were not successful. Because the toxic action of pheromones upon plant tissues appears relatively benign, and growers have not been adverse to localized phytotoxicity to foliage and fruits on two trees per 0.4 ha, we propose that limited phytotoxicity associated with first-generation aerosol dispenser technology can be viewed as nonthreatening.

A molecular method is presented for differentiating the morphologically cryptic leafminers Liriomyza langei Frick and L. huidobrensis (Blanchard). This method requires polymerase chain reaction (PCR) amplification of a 1031-bp region of mitochondrial cytochrome oxidase DNA followed by restriction fragment analysis using the restriction enzymes SpeI and EcoRV. SpeI cuts the mitochondrial fragment of L. langei into two fragments, but does not cut the L. huidobrensis fragment. EcoRV cuts the L. huidobrensis fragment into two fragments, but does not cut the L. langei fragment. This PCR-restriction fragment-length polymorphism (RFLP) method is faster and less costly than DNA sequencing, which is currently the only other way to differentiate these two species. We apply the method to samples from recently introduced leafminer populations in Sri Lanka, Canada, and South Africa and find that the invasive leafminer in all three locations is L. huidobrensis.

Studies examined the possibility that mixtures of endophytic perennial ryegrass and Kentucky bluegrass provide resistance against larvae of the black cutworm, Agrotis ipsilon (Hufnagel). We hypothesized that resistance against A. ipsilon in such stands would stem from the influence of Kentucky bluegrass on A. ipsilon growth and behavior rather than the influence of endophytic perennial ryegrass. In replicated greenhouse experiments, black cutworm larvae initially emigrated more quickly from pots containing monocultures of endophytic perennial ryegrass than from Kentucky bluegrass monocultures or polycultures of Kentucky bluegrass and endophytic perennial ryegrass. However, biomass of emigrating larvae decreased linearly as the proportion of Kentucky bluegrass increased. Turfgrass mixtures containing endophytic perennial ryegrass and Kentucky bluegrass may provide resistance against A. ipsilon mainly through the physiological effects of Kentucky bluegrass on A. ipsilon growth and development, but possibly through the influence of endophytic perennial ryegrass on A. ipsilon movement and foraging behavior as well.

Research on consumer, grower, and landscape manager perception of azalea lace bug, Stephanitis pyrioides (Scott), feeding and on plant productivity parameters, including gas exchange and growth, has increased our understanding of the nature of feeding injury. These studies made it possible to develop decision-making guidelines for cost-effective maintenance of aesthetically pleasing azaleas. Criteria were considered for three management situations: a 0.41-ha (1-acre) nursery production system that may use either insecticidal soap, acephate, or imidacloprid to control lace bugs; a landscape planting of a group of 10 azaleas; or maintenance of a single azalea in the landscape. Lace bug thresholds were calculated using a hybrid economic injury level (EIL) formula. Pesticide application decisions were determined using survey-based data from grower, landscape manager, and consumer perceptions of unacceptably injured azaleas at point-of-purchase for the nursery situation. Additional landscape scenarios incorporated the perceptions of growers, landscape managers, and consumers for those levels of lace bug feeding-injury that prompted the desire for treatment. Hybrid EIL determinations are appropriate for lace bug management in landscape systems where landscape professionals manage large plantings of azaleas and as a component of pest management among nursery production systems. Aesthetic considerations are more appropriate in determining control thresholds among a few or individual azaleas in the landscape.

Foraging behavior of the Formosan subterranean termite, Coptotermes formosanus Shiraki, was studied in the laboratory by using two-dimensional foraging arenas containing multiple foraging sites. Within each arena, 16 foraging sites were arranged in a uniform grid pattern and foragers were introduced into the arena through a central initiation site. Chi-square analysis determined the frequency of tunnels was uniformly distributed around the perimeter of the initiation site but became significantly skewed toward the foraging sites at a distance where the foraging sites could be encountered. Tunnel distribution was similar whether wood was present or absent at the foraging sites, suggesting that foragers respond to structural anomalies in the substrate rather than simply to the presence of food. Also described is the generalized sequence of events as foragers tunnel throughout the arenas.

The effects of four plant extract formulations on the orientation and survival of subterranean termites were tested. In choice experiments, extract-treated filter paper had a significantly repellent effect on groups of Reticulitermes santonensis De Feytaud, R. virginicus (Banks), Coptotermes formosanus Shiraki, and Schedorhinotermes intermedius Breinli. There was no species-specific difference in avoidance behavior toward the tested concentrations. No-choice experiments revealed toxic properties of all investigated extracts by contact or airborne compounds against R. santonensis. However, high termite mortality was only achieved by forced direct or forced indirect exposure to the plant material. Feeding deterrence or digestive toxicity could not be judged in these experiments. One of the extracts was efficiently used for soil treatments to protect a food substrate against R. santonensis infestation. Extract-treated barriers in the experiments did not affect mortality compared with control trials but prevented termites from penetrating treated soil.

Effects of the synergists piperonyl butoxide (PBO) and S,S,S-tributyl phosphorotrithioate (DEF) on propoxur pharmacokinetics were examined in the German cockroach, Blattella germanica (L.). Treatment of adult male German cockroaches with the cytochrome P450 monooxygenase inhibitor, PBO, or the esterase inhibitor, DEF, increased propoxur toxicity by 2- and 6.8-fold, respectively, implicating hydrolysis as a major detoxification route of propoxur in the German cockroach. However, significant hydrolytic metabolism could not be demonstrated conclusively in vitro resulting in a conflict between in situ bioassay data and in vitro metabolic studies. In vitro propoxur metabolism with NADPH-fortified microsomes produced at least nine metabolites. Formation of metabolites was NADPH-dependent; no quantifiable metabolism was detected with cytosolic fractions. However, microsomal fractions lacking an NADPH source did produce a low, but detectable, quantity of metabolites (1.6 pmol). PBO inhibited NADPH-dependent propoxur metabolism in a dose-dependent fashion, implicating cytochrome P450 monooxygenases as the enzyme system responsible for the metabolism. Interestingly, DEF also inhibited the NADPH-dependent metabolism of propoxur, albeit to a lower extent. Treatment with PBO or DEF also caused a significant reduction in the cuticular penetration rate of propoxur. The data demonstrate that unanticipated effects are possible with synergists and that caution must be exercised when interpreting synergist results.

Lethal time to mortality responses were established for eight insecticides against workers and soldiers of the Formosan subterranean termite, Coptotermes formosanus Shiraki, and workers of Reticulitermes virginicus (Banks). There were significant differences in the tolerance ratios between workers of C. formosanus colonies to all toxicants tested except fipronil. One colony was 16 times more tolerant than another to deltamethrin. C. formosanus soldiers had significant differences in tolerance ratios among colonies exposed to all toxicants except chlorpyrifos. Methoxychlor, permethrin, deltamethrin, and fipronil did not kill soldiers from two, one, one, and three colonies, respectively, within 8 h. Seventy-five percent of R. virginicus colonies were significantly less susceptible than the most susceptible colony to chlordane, methoxychlor, chlorpyrifos, cypermethrin, and fipronil, with 50% of the colonies less susceptible to permethrin and bendiocarb. In 50% of C. formosanus colonies the worker lethal time curves displayed substantial flattening in response to permethrin, and deltamethrin. Lethal time curves for C. formosanus soldiers exposed to chlordane, chlorpyrifos, permethrin, cypermethrin, deltamethrin, and bendiocarb showed substantial flattening. R. virginicus workers demonstrated substantial curve flattening when exposed to chlordane, methoxychlor, chlorpyrifos, deltamethrin, and fipronil. These findings indicate substantial inter-colony and intra-colony differences in susceptibility to insecticides.

Foraging cockroaches ingest insecticide baits, translocate them, and can cause mortality in untreated cockroaches that contact the foragers or ingest their excretions. Translocation of eight ingested baits by adult male Blattella germanica (L.) was examined in relation to the type of the active ingredient, formulation, and foraging area. Ingested boric acid, chlorpyrifos, fipronil, and hydramethylnon that were excreted by adults in small dishes killed 100% of first instars within 10 d and >50% of second instars within 14 d. Residues from these ingested baits were also highly effective on nymphs in larger arenas and killed 16–100% of the adults. However, when the baits and dead cockroaches were removed from the large arenas and replaced with new cockroaches, only residues of the slow-acting hydramethylnon killed most of the nymphs and adults, whereas residues of fast acting insecticides (chlorpyrifos and fipronil) killed fewer nymphs and adults. Excretions from cockroaches that ingested abamectin baits failed to cause significant mortality in cockroaches that contacted the residues. These results suggest that hydramethylnon is highly effective in these assays because cockroaches that feed on the bait have ample time to return to their shelter and defecate insecticide-laden feces. The relatively high concentration of hydramethylnon in the bait (2.15%) and its apparent stability in the digestive tract and feces probably contribute to the efficacy of hydramethylnon. To control for differences among baits in inert ingredients and the amount of active ingredient, we compared 1% chlorpyrifos with 1% hydramethylnon in identical baits. Again, hydramethylnon residues provided greater secondary kill, but the results highlighted the importance of the inert ingredients. We conclude that, in the absence of cannibalism and necrophagy, translocation of baits and secondary kill are most effective with slow acting insecticides in palatable baits that can traverse the digestive tract and be deposited within and around the cockroach aggregation.

We evaluated the effects of Bacillus thuringiensis (Bt) toxin Cry1Ac on survival and development of a susceptible strain and laboratory-selected resistant strains of pink bollworm, Pectinophora gossypiella (Saunders). For susceptible and resistant strains tested on artificial diet, increases in Cry1Ac concentration reduced developmental rate and pupal weight. In greenhouse tests, survival of resistant larvae on transgenic cotton that produces Cry1Ac (Bt cotton) was 46% relative to their survival on non-Bt cotton. In contrast, Bt cotton killed all susceptible larvae tested. F₁ hybrid progeny of resistant and susceptible adults did not survive on Bt cotton, which indicates recessive inheritance of resistance. Compared with resistant or susceptible larvae reared on non-Bt cotton, resistant larvae reared on Bt cotton had lower survival and slower development, and achieved lower pupal weight and fecundity. Recessive resistance to Bt cotton is consistent with one of the basic assumptions of the refuge strategy for delaying resistance to Bt cotton. Whereas slower development of resistant insects on Bt cotton could increase the probability of mating between resistant adults and accelerate resistance, negative effects of Bt cotton on the survival and development of resistant larvae could delay evolution of resistance.

Inheritance of the high-level diflubenzuron resistance shown by a laboratory-selected strain of Lucilia cuprina (Wiedemann) was examined in matings with a susceptible reference strain. Progeny of reciprocal crosses between resistant females and susceptible males showed higher LC₅₀ values than the alternate reciprocal cross, indicating some maternal influence on inheritance of resistance. Resistance was inherited in a codominant (S♂ × R♀) or incompletely recessive (R ♂ × S♀) manner. Monooxygenase activities (aldrin epoxidation) of the F₁ generations were also intermediate between the levels shown by the parental lines, however, inheritance of enzyme activities showed greater degrees of dominance than for resistance levels. There was also some maternal influence on inheritance of monooxygenase activities. Backcrosses of F₁ generations to both susceptible and resistant parents did not fit the expected patterns for a major sex-linked resistance locus, indicating that the maternal influence on resistance inheritance was not associated with sex-linkage of a major resistance gene. The backcross data also failed to fit the model for a single major autosomal gene, suggesting that the resistance in the diflubenzuron-selected strain is polygenic, involving mechanisms additional to monooxygenases.

The sesquiterpene carboxylic acids (SCA), ( )-(E)-α-santalen-12-oic, (−)-(E)-endo-α-bergamoten-12-oic, and ( )-(E)-endo-β-bergamoten-12-oic acid, are produced in glandular trichomes of Lycopersicon hirsutum f. typicum Humb. & Bonpl. accession (LA) 1777, which is highly resistant to a range of pests of cultivated tomatoes, L. esculentum Mill. Exposure of the larvae of two key tomato pests, tomato fruitworm [Helicoverpa zea (Boddie)] and beet armyworm [Spodoptera exigua (Hübner)], to these compounds in diets and on leaf surfaces resulted in reduced development rates and survival and deterred feeding. These effects were observed when levels of SCA, in artificial insect diet, applied to leaflets of susceptible cultivars, or synthesized in trichomes of leaves of plants, exceeded 2 mg SCA/g of diet or fresh leaf weight. This study suggests that cultivated tomatoes capable of synthesizing SCA, at 2 mg SCA/g of leaf tissue or greater, on their leaves and fruit would display enhanced host plant resistance to H. zea and S. exigua and other insect pests.

We recently described a new source of host-plant resistance to the Colorado potato beetle, Leptinotarsa decemlineata (Say), in a tetraploid potato (Solanum tuberosum L.) selection, ND2858–1. This genotype, and selected backcross progeny, had little damage while check cultivars were defoliated in open-choice field assays. To further characterize the observed deterrence, we determined foliar glycoalkaloids and conducted no-choice assays with ND2858–1 backcross progeny genotypes (ND4382-n). Development of neonate L. decemlineata in detached leaf assays on resistant progeny genotypes was delayed and larval weight gain after 4 d was inhibited by 75% relative to larval development and weight gain on susceptible genotypes. Inhibition of larval development in detached leaf assays with the selected progeny genotypes was equivalent to that of high-leptine genotypes of S. chacoense Bitter. Foliar glycoalkaloids of resistant genotypes included low levels of leptines I and II. The unlikely nature of this cross and the presence of leptine in this and resistant progeny selections cast doubt on the recorded pedigree. Molecular analyses were conducted by restriction fragment-length polymorphism and amplified fragment-length polymorphisms. Both methods established a high degree of relatedness to S. tuberosum and S. chacoense but not to S. fendleri. We conclude that ND2858–1 did not originate from a cross with S. fendleri, but is likely derived from S. chacoense. Oviposition and larval survival were reduced when adult L. decemlineata were placed in cages with resistant genotypes; an effect that was enhanced by inclusion of Perillus bioculatus F. Therefore, the nonpreference previously observed in open-choice field defoliation assays is also associated with antibiotic effects on L. decemlineata. The resistance may be caused by leptines, but is greater than would be expected by the leptine content. This source of host plant resistance could be a cost-effective management strategy, especially if combined with other resistance mechanisms or compatible control measures to delay development of resistance in the target insects.

Transgenic varieties of field corn that express the Cry1Ab B. thuringiensis (Bt) toxin in ear tissue present the potential of reducing ear feeding by the corn earworm, Helicoverpa zea (Lepidoptera: Noctuidae), and for reducing the size of populations of the insect infesting other host crops. Life history parameters of H. zea feeding on ears of conventional and Bt field corn varieties were measured in field plots in eastern North Carolina in 1997 and 1998. Transformation events investigated were Mon-810 and Bt-11. Bt corn was found to cause a steady mortality of larvae during development, but permitted ≈15–40% survival to the prepupal stage compared with non-Bt corn. Mortality of prepupae and pupae from Bt corn was also higher than from non-Bt corn, reducing overall adult production by 65–95%. The larvae that did survive grew more slowly on Bt than on non-Bt corn, and produced pupae that weighed 33% less. Pupation and adult eclosion were delayed by 6–10 d by feeding on Bt corn ears. Corn varieties expressing Bt in ear tissue have the potential to reduce H. zea ear feeding by up to 80%, and the potential to reduce populations emerging from ear-stage corn fields to infest cotton, soybean and other crops by around 75%. To have a measurable effect on area-wide populations, Bt corn varieties would need to be planted in large proportions of corn fields. Extensive planting of varieties such as those tested here, having only moderate effects on H. zea, would raise concerns about rapid evolution of resistance.

Griffonia simplicifolia lectin II (GSII) is a plant defensive protein that significantly delays development of the cowpea bruchid Callosobruchus maculatus (F.). Previous structure/function analysis by site-directed mutagenesis indicated that carbohydrate binding and resistance to insect gut proteolysis are required for the anti-insect activity of this lectin. However, whether there is a causal link between carbohydrate binding and resistance to insect metabolism remains unknown. Two proteases principally responsible for digestive proteolysis in third and fourth instar larvae of C. maculatus were purified by activated thiol sepharose chromatography and resolved as cathepsin L-like proteases, based on N-terminal amino acid sequence analysis. Digestion of bacterially expressed recombinant GSII (rGSII) and its mutant protein variants with the purified gut proteases indicates that carbohydrate binding, presumably to a target ligand in insect gut, and proteolytic resistance are independent properties of rGSII, and that both facilitate its efficacy as a plant defensive molecule.

The effects of nitrogen fertilizer on sweet potato, Ipomoea batatas (L.) Poir., resistance to the sweetpotato weevil, Cylas formicarius elegantulus (Summers), was studied. Adult weevil feeding and oviposition preference, larval survival, and pupal weight were used as measures of sweet potato resistance. Sweet potato resin glycosides and caffeic acid concentrations in the periderm tissue of storage roots also were measured. Sweet potato genotypes (Beauregard, Excel, W-244, W-250, and Sumor) with varying levels of resistance to sweetpotato weevil were grown in the field under three nitrogen regimes (0, 45, and 135 kg N/ha). Harvested storage roots were evaluated in the laboratory for feeding and oviposition activity of sweetpotato weevil female adults under no-choice and choice test conditions. Larval survival rate and pupal weight were determined by rearing the insects individually on storage root sections. Nitrogen level had a significant effect on the number of eggs deposited, but not on the number of feeding punctures. Sweetpotato weevils laid fewer eggs on plants with the highest level of nitrogen. Nitrogen levels did not significantly affect larval survival and pupal weight. Genotype had a significant effect on feeding, oviposition, and larval survival. Beauregard had higher levels of feeding, oviposition, and larval survival compared with the other genotypes. No interaction effects between nitrogen and genotype were significant. Resin glycosides and caffeic acid concentrations were significantly different among genotypes and between years. Nitrogen levels significantly affected the concentrations of caffeic acid in 1997.

We determined whether immersion in 49°C water for 20 min, a quarantine treatment developed for disinfestation of fruit flies in lychee, Litchi chinensis Sonn., and longan, Dimocarpus longan (Lourd.) Steud., exported from Hawaii, would also disinfest fruit of two species of Cryptophlebia. The pattern of tolerance to heat in Cryptophlebia illepida (Butler) was generally eggs < neonates < early instars = late instars < pupae. No C. illepida survived immersion for 16 or 20 min. Late fourth and fifth instars were determined to be the most tolerant stage that occurs in harvested fruit. Late instars of Cryptophlebia ombrodelta (Lower) were more tolerant of hot-water immersion than those of C. illepida, but no C. ombrodelta late instars survived immersion for 16 or 20 min. The hot water immersion quarantine treatment for fruit flies should effectively disinfest lychees and longans of any Cryptophlebia.

Six plant essential oils alone as repellent and fumigant, and in combination with the controlled atmosphere against Liposcelis bostrychophila Badonnel were assessed in the laboratory. These essential oils were extracted from the leaves of six source plants: Citrus tangerina Tanaka, Citrus aurantium L., Citrus bergamia Risso et Poiteau, Pinus sylvestris L., Cupressus funebris Endl., and Eucalyptus citriodora Hook. The repellency test indicated that L. bostrychophila adults were repelled by filter paper strips treated with six essential oils. Of these essential oils, the C. funebris oil was most effective followed by that of P. sylvestris, C. tangerina, C. bergamia, and E. citriodora. The average repellency of the C. aurantium oil against L. bostrychophila adults was significantly lower than other five test oils by day 14. These essential oils had a high level of toxicity in the fumigation assay against L. bostrychophila adults at both 10 and 20 ppm. When combined with two controlled atmosphere treatments (12% CO₂ 9% O₂, and 10% CO₂ 5% O₂, balanced N₂), the toxicity of plant oils was enhanced significantly.

The prospects of using low pressure that creates a low-oxygen atmosphere to control stored-product insects were investigated in the laboratory. Eggs, larvae, and pupae of Tribolium castaneum (Herbst), Plodia interpunctella (Hübner), and Rhyzopertha dominica (F.) were exposed to 32.5 mmHg in glass chambers at 25, 33, 37, and 40°C for times ranging from 30 min to 144 h. Time-mortality data were subjected to probit analyses and lethal dose ratios were computed to determine differences in lethal time (LT) values among all species-life stage combinations across the four temperatures. Eggs of each species were the life stage most tolerant to low pressure. Pupae of T. castaneum and R. dominica were more tolerant to low pressure than larvae. In all life stages, mortality increased with increasing exposure time to low pressure and also with increasing temperature. Immature stages of R. dominica were more tolerant to low pressure than immature stages of the other two species. The LT₉₉ for R. dominica eggs was 176.32 h at 25°C and that for P. interpunctella eggs was 28.35 h at the same temperature. An increase in temperature to 33°C resulted in a LT₉₉ of 85.98 h for R. dominica and 6.21 h for P. interpunctella. Higher temperatures resulted in further significant reduction in lethal time values. Low pressure represents a simple, nonchemical alternative to fumigants such as methyl bromide and phosphine for controlling pests of stored-products or other commodities.

A sand dilution assay was developed to study how composting affects the nutritional value of stored laying hen manure for larvae of the house fly, Musca domestica L. Equal numbers of eggs were inoculated into graded amounts of stock manure and incubated under standardized moisture conditions. Survival and mass per emerging adult diminished with progressively lower supplies of manure per larva, whether the manure was diluted into clean, white sand or placed on top of an equal volume of sand. Mass of adults per original egg was an increasing linear function of log₂ manure supply, with extrapolated lower supply threshold, S_L = 0.06 g per egg. It is proposed that S_L is a measure of a substrate’s nutritional value—the greater the threshold, the lower its value. Dilution of the same stock manure in loam or sandy loam reduced the manure’s apparent nutritional value, and dehydration of the stock manure to 20% water before rehydration to 70% also reduced nutritional value. Assays of bulk samples from replicated piles of laying hen manure mixed with sunflower hulls indicated the mixture was nutritionally equivalent to the stock manure, but that 3–4 wk of subsequent aerobic, thermophilic composting reduced it to ≈10% of its initial value. These results suggest that composting may be a useful technique for reducing the fly breeding potential of laying hen manure and other substrates that must be stored before spreading and incorporation on crop land.
