To gauge the impact of COVID-19 on the Canadian beekeeping sector, we conducted a survey of over 200 beekeepers in the fall of 2020. Our survey results show Canadian beekeepers faced two major challenges: 1) disrupted importation of honey bees (Hymenoptera: Apidae) (queen and bulk bees) that maintain populations; and 2) disrupted arrival of temporary foreign workers (TFWs). Disruptions in the arrival of bees and labor resulted in fewer colonies and less colony management, culminating in higher costs and lower productivity. Using the survey data, we develop a profitability analysis to estimate the impact of these disruptions on colony profit. Our results suggest that a disruption in either foreign worker or bee arrival allows beekeepers to compensate and while colony profits are lower, they remain positive. When both honey bee and foreign workers arrivals are disrupted for a beekeeper, even when the beekeeper experiences less significant colony health and cost impacts, a colony with a single pollination contract is no longer profitable, and a colony with two pollination contracts has significantly reduced profitability. As COVID-19 disruptions from 2020 and into 2021 become more significant to long-term colony health and more costly to a beekeeping operation, economic losses could threaten the industry's viability as well as the sustainability of pollination-dependent crop sectors across the country.The economic and agricultural impacts from the COVID-19 pandemic have exposed a vulnerability within Canada's beekeeping industry stemming from its dependency on imported labor and bees.Travel disruptions and border closures pose an ongoing threat to Canadian agriculture and apiculture in 2021 and highlight the need for Canada's beekeeping industry to strengthen domestic supply chains to minimize future risks.

Neoseiulus cucumeris (Oudemans) (Acari: Phytoseiidae), an economically important biocontrol agent, was reared on almond pollen for 50 generations. We evaluated the effect of rearing predators on this pollen by comparing biological parameters at different generations (G1–G50). The shortest and longest development time occurred in G5 and G10, respectively. Females at the fifth and 50th generations laid eggs earlier than those of other generations. Females at G50 laid eggs over a longer period and produced more eggs than females of other generations, although females in the earlier generations had a higher gross reproductive rate and net reproductive rate than later generations. The intrinsic rate of increase, as well as the finite rate of increase of N. cucumeris in the fifth and 50th generations was significantly greater than those in other generations, while the first generation had the lowest values of these parameters. The dorsal shield length of both females and males and the width of females were found to be unaffected by their constant feeding on almond pollen. However, the number of rearing generations significantly affected the width of males. Long-term rearing of N. cucumeris for at least 50 generation on almond pollen did not substantially affect the predator's quality and this food source could be used for the mass production of this predator. Almond pollen should be assessed in rearing other phytoseiid mites that are important in biocontrol strategies.

Pathogenic assessment of a baculovirus-based biopesticide containing Spodoptera frugiperda multiple nucleopolyhedrovirus (SfMNPV: Baculoviridae: Alphabaculovirus) infecting fall armyworm, Spodoptera frugiperda (J. E. Smith, 1797) (Lepidoptera: Noctuidae) is reported. In the bioassays, neonates were infected with different doses of SfMNPV applied on Cry1Ac Bt soybean and non-Bt soybean. Our findings indicated that S. frugiperda neonates did not survive at 10 d post infection or develop into adults on Bt and non-Bt soybean sprayed with the field recommended dose of SfMNPV. In contrast, a proportion of the infected neonates developed into adults when infected with lower doses of SfMNPV (50%, 25%, and 10% of field dose) in both Bt and non-Bt soybean. However, S. frugiperda neonates surviving infection at the lowest virus doses on both soybean varieties showed longer neonate-to-pupa and neonate-to-adult periods, lower larval and pupal weights, reduced fecundity, and increased population suppression. Nevertheless, more pronounced pathogenicity of SfMNPV infecting neonates of S. frugiperda were verified on larvae that developed on Bt soybean. These findings revealed that, beyond mortality, the biopesticide containing SfMNPV also causes significant sublethal pathogenic effects on neonates of S. frugiperda developing on Bt and non-Bt soybean and suggested an additive effect among SfMNPV and Cry1Ac insecticidal protein expressed in Bt soybean.

In this research, functional responses of Amblyseius swirskii Athias-Henriot preying on different Tetranychus urticae Koch nymphal densities (2, 4, 8, 16, 32, 64, and 128) were studied at eight constant temperatures (15, 20, 25, 27.5, 30, 32.5, 35 and 37.5°C) in a circular Petri dish (3-cm diameter × 1-cm height) under lab conditions. At all temperatures, the logistic regression showed a type II functional response. A nonlinear relationship was found between temperature and attack rate and the reciprocal of handling time. The reciprocal of handling time decreased exponentially with increasing temperature. In contrast, the attack rate grew rapidly with increasing temperatures up to an optimum, showing a decreasing trend at higher temperatures. In order to quantify the functional response of A. swirskii over a broad range of temperatures and to gain a better estimation of attack rate and handling time, a temperature-settled functional response equation was suited to our data. Our model showed that the number of prey consumed increased with rising prey density. Also, the predation rates increased with increasing temperatures but decreased at extremely high temperatures. Based on our model, the predation rate begins at the lower temperature threshold (11.73°C) and reaches its peak at upper temperature threshold (29.43°C). The coefficient of determination (R²) of the random predator model was 0.99 for all temperatures. The capability of A. swirskii to search and consume T. urticae over a wide range of temperatures makes it a good agent for natural control of T. urticae in greenhouses.

Studies were conducted with ozone gas fumigation under vacuum as a methyl bromide alternative against life stages of coffee berry borer (CBB) Hypothenemus hampei (Ferrari) (Coleoptera: Curculionidae: Scolytinae), and the urediniospores of coffee leaf rust (CLR), Hemileia vastatrix Berkeley & Broome (Basidiomycota: Pucciniales) in green coffee, Coffea spp. L. Fumigation with 10,000 ppm O₃ gas under –25.4 mm Hg vacuum1 at 13.0 ± 3.0°C for 6.0 h killed all CBB larvae, pupae, and adults, but did not kill all CBB eggs (∼15% survival). Mortality of CLR urediniospores was 100% within the first hour of the 6-h fumigation. Ozone fumigation had no adverse effects on coffee quality. Results indicated that CBB adult hitchhikers may be the only target life stage of quarantine concern, and additional studies focused on this stage. CBB adult survival and reproduction decreased significantly at moisture contents ≤20%, and F₁ generation survival did not occur in green coffee at moisture contents ≤15%. As the international standard for green coffee moisture content is 9–12%, adult CBB should not survive or reproduce in exported dry green coffee. Standard industry processing of harvested coffee cherries to the green coffee stage using either mechanical- or sun-drying eliminated CBB infestations from the field. A systems approach is recommended for exporting green coffee to control CBB and CLR that includes eliminating CBB life stages with standard processing methods, reducing moisture content to 9–12% to prevent egg deposition, survival or reproduction, and O₃ fumigation to ensure quarantine security against potential CBB adult hitchhikers.

Ethyl formate (EF) was evaluated as a potential alternative to methyl bromide (MB) for phytosanitary treatment of imported citrus fruit in the Republic of Korea. Planococcus citri (Risso) (Hemiptera: Pseudococcidae), a mealybug with known tolerance against EF and MB, was used as a representative pest to test efficacy of the two fumigants against eggs. In nine commercial-scale refrigerated container (67.5 m³) trials using imported orange, lemon and grapefruit, EF applied at the currently approved dose for citrus (70 g·m^-3 at 5°C for 4 h, developed for Aspidiotus excisus Green (Hemiptera: Diaspididae), a species less EF tolerant than P. citri) resulted in 76.9–98.3% mortality of P. citri eggs. The EF treatment did not affect the sugar content or the color of peel and pulp of the treated fruit. When oranges were treated according to the current MB (64 g·m^-3 at >5°C for 2 h) or EF treatment guidelines, the concentration of fumigant around the fruit fluctuated between 9.4 and 185.1 ppm for EF and 9.5–203.0 ppm for MB during the 72-h post-fumigation processes (venting [0–2 h], transportation to storage [2–24 h], and storage periods [24–72 h]) with both EF and MB maintained between 10 and 100 ppm during the storage period. Considering the efficacy of EF, its apparent lack of phytotoxicity, and its more manageable threshold limit value for humans (100 ppm EF compared to 1 ppm MB for an 8-h time weighted average exposure), our results suggest that EF may be a promising alternative to MB for the phytosanitary treatment of imported citrus in Korea.

Brown marmorated stink bug (BMSB), Halyomorpha halys (Stål) (Hemiptera: Pentatomidae), is a pest of concern that must be controlled for market access of host material and regulated articles to certain countries. This work outlines a rearing system for BMSB on live cowpea plants, Vigna unguiculata (L.) Walp. (Fabales: Fabaceae), including methods to induce adults to both enter and exit diapause. This scalable system affords continuous access to >600 specimens per week of each life stage and/or age group, which is particularly advantageous when developing treatment efficacy data.

The Mediterranean fruit fly, Ceratitis capitata (Wiedemann), is one of the most important pests of fruits worldwide. In the present study, we investigated the ovipositional response of C. capitata females to 37 compounds of fruit volatiles from various chemical groups and the dose response to five of them. Red plastic hollow hemispheres (domes) were used as oviposition substrates in all tests. Twenty of the compounds tested increased female egg laying compared to control substrates that contain no chemicals. With 16 compounds, similar number of eggs was deposited in treated and in control oviposition substrates. One terpene ((±)-linalool) reduced egg laying indicating a deterrent ovipositional effect. Both the esters and aldehydes tested increased the ovipositional responses in C. capitata. Most of the monoterpene hydrocarbons increased oviposition, while oxygenated monoterpenes, sesquiterpenes hydrocarbons, and oxygenated sesquiterpenes had mostly neutral effect. Ethyl hexanoate and R-(+)-limonene increased oviposition in the majority of doses tested. Different doses of (–)-linalool elicited differential female ovipositional responses. In contrast, valencene and citral, regardless of dose did not affect female oviposition. Practical implications of these findings are discussed.

Chysodeixis includens (Walker) is a polyphagous economic pest in agricultural landscapes. To detect the occurrence of this pest in the field, trapping using sex pheromone lures is often implemented. However, other plusiine species are cross-attracted to these lures and may be misidentified as C. includens due to their morphological similarities. The objectives of this study were to provide region-specific information on the abundance of C. includens throughout the year as well as document the occurrence of related plusiines cross-attracted to C. includens sex pheromone traps in the Florida Panhandle. Twelve commercial fields of peanut and twelve commercial fields of cotton located across Escambia, Santa Rosa, and Jackson counties were monitored with Trécé delta traps baited with C. includens sex pheromone lures (Alpha Scents, Inc.; West Linn, OR) from June 2017 to June 2019. There was no difference in C. includens flight across dryland or irrigated fields. Identifications revealed that in addition to C. includens, the following species of the subfamily Plusiinae were crossed-attracted: Argyrogramma verruca (Fabricius) (Lepidoptera: Noctuidae), Trichoplusia ni (Hübner) (Lepidoptera: Noctuidae), Ctenoplusia oxygramma (Geyer) (Lepidoptera: Noctuidae), and Rachiplusia ou (Guenée) (Lepidoptera: Noctuidae). The occurrence of each species in the region and their flight phenology are documented. Chysodeixis includens abundance was greatest in September and decreased through December. Due to the high abundance of C. oxygramma and similar flight phenology to C. includens, this is the likeliest species to skew estimations and influence management decisions of C. includens, especially early in the crop season, when C. includens abundance is low.

The bean bug, Riptortus pedestris (Fabricius), is a serious pest of legume crops in East Asia. Here, we report how the consumption of different types of legume seeds affects various aspects of nymphal and adult traits related to fitness in R. pedestris. Two experiments were conducted. In the first experiment, we assessed how R. pedestris nymphs and adults performed on one of 10 different legume seeds: adzuki bean, chickpea, cowpea, two kidney bean varieties (red, white), mung bean, peanut, small black bean, and two soybean varieties (black, yellow). Riptortus pedestris fed on yellow soybean and chickpea performed well in terms of nymphal survivorship and fecundity. However, R. pedestris fed on two kidney bean varieties suffered 100% nymphal mortality and reduced fecundity. Small black bean and black soybean supported high fecundity, but were not suitable for nymphal development. Lipid content was 4- to 11-folds higher in R. pedestris raised on peanuts than those on the others. In the second experiment, we recorded the key parameters of adult performance from R. pedestris assigned to one of nine combinations of three nymphal and three adult diets (yellow soybean, adzuki bean, peanut). Riptortus pedestris raised on yellow soybean during development produced more eggs over their lifetime than those raised on the others. However, this beneficial effect of consuming yellow soybean during development occurred only when yellow soybean was consumed during adulthood. Our data have implications for predicting the occurrence and population dynamics of this pest.

Diuraphis noxia, Russian wheat aphid (Hemiptera: Aphididae), established in Australia since 2016, is dependent on grasses (Poales: Poaceae) to persist in the low-rainfall Australian wheat belt, where no crops are present during summer. To identify grasses as D. noxia hosts in Australia, plants were tested in greenhouse conditions as either whole plants collected from roadsides or grown from collected seed in 2017 and 2018. To determine actual field refugia, direct grass sampling and Berlese extraction of aphids were conducted from October 2018 to May 2020 throughout Southern Australia (2,285 samples). One hundred and twenty-six grass species were collected, 54 showed presence of D. noxia, of which 24 were considered host plants, including 16 species (9 Australian natives) not recorded as host plants previously. Hordeum leporinum (Link) Arcang. Poales:Poaceae and several Bromus species (Poales: Poaceae) showed the highest D. noxia detection frequency and aphid numbers, but these introduced grass species are not summer active in most of South Australia. The native Enneapogon nigricans (Poales: Poaceae) (R.Br.) is the most important summer refuge species because of its widespread distribution, summer growth, and an intermediate level of positive detections with low D. noxia populations. The late summer represents the main bottleneck for D. noxia with very few hosts available and very low D. noxia detections overall. Late summer rainfall (February) seems essential to have the main host grasses germinate for D. noxia populations to build up and potentially invade crops sown in autumn.

Euschistus heros (Hemiptera: Pentatomidae) has two allopatric strains with a hybrid zone in central Brazil. Asymmetric dispersal and gene flow between these strains of E. heros have been observed, where the South strain (SS) moves more quickly to the northern regions of the country than the North strain (NS) to the southern areas. In addition, SS generally has a bigger body size and presents dark brown coloration, and NS is usually smaller in size and presents light brown coloration. Here, we studied the reproductive behavior and tested for the presence of assortative mating and reproductive barriers between the two allopatric strains of E. heros. Nonrandom mating was observed in the SS strain based on mating choice trials and the reproductive isolation indexes. SS females and males prefer to mate with their co-specific (same strain) partner, while NS insects showed no mating preference. The insect's pronotum width was positively associated with the mating choice suggesting size-assortative mating in E. heros. Reciprocal crosses between strains yielded similar reproductive outputs when compared with pure strain crosses, suggesting similar fitness of hybrid pure strains. The asymmetric gene flow in the hybridization zone that favors SS seems to be associated with the reproductive behavior of the species, which favors the typical phenotype found in the SS populations.

In order to investigate the effect of Wolbachia on fitness of their hosts, the biological characteristics of a primarily parthenogenetic line and a bisexual restored line (obtained by antibiotic treatment) of Trichogramma pretiosum were compared in the laboratory. Results indicated that both the mean longevity and fecundity of parthenogenetic line (14.2 d and 165 eggs/female) were significantly higher than those in bisexual restored line (8.4 d and 124.2 eggs/female). Both lines of T. pretiosum had the highest daily fecundity (23.3 eggs/female for parthenogenetic line and 19.8 eggs/female for bisexual restored line) on the first day during their reproduction period, and their survival rate and daily fecundity decreased gradually with age; however, the survival rate and daily fecundity of the parthenogenetic line were always higher than those of bisexual restored line, during the entire experimental period. There was no significant difference in emergence rate and deformity rate between the two T. pretiosum lines. The life-table parameter results indicated that net reproduction rate (R₀) and mean generation time (T) in parthenogenetic line (133 and 16.8) were significantly higher than those in bisexual restored line (61.7 and 15.5); intrinsic rate of natural increase (r_m) and finite rate of increase (λ) in parthenogenetic line (0.29 and 1.34) were higher than those in bisexual restored line (0.26 and 1.3). Based on these results, it can be confirmed that the removal of Wolbachia in the parthenogenetic line had an adverse effect on fitness of T. pretiosum.

Olfaction is of great significance for insect mate-seeking and host-locating behaviors. Insect odorant-binding proteins (OBPs), especially those antenna-enriched OBPs, are thought to discriminate, capture and transport odorant molecules to olfactory receptors, but this has not been fully clarified in Bactrocera minax (Enderlein), an economically important pest of citrus crops. Our previous studies showed that seven OBP genes (BminOBP1-7) were identified from B. minax adults via a head transcriptome analysis, of which only BminOBP3 and 6 were highly expressed in antennae, suggesting an olfactory role. To confirm their functions, here, BminOBP3 and 6 were cloned, expressed in Escherichia coli cells. Binding properties of the recombinant BminOBPs with 13 volatiles, most of which can elicit a significant behavioral response from B. minax adults, were determined by fluorescent competitive binding assays. The results showed that Both BminOBP3 and 6 exhibited a remarkable selectivity towards the 13 ligands tested. BminOBP3 displayed strong binding affinity only with undecanol. BminOBP6 demonstrated strong binding affinity with undecanol and limonene among 13 ligands tested. Undecanol is believed to be main sex pheromone component of B. minax. Limonene is an important volatile compound enriched in citrus fruits. Taken together, we concluded that BminOBP3 and 6 may play a prominent role in the process of B. minax mate-seeking and host-locating behaviors through recognizing and transporting these volatiles. It is conceivable that this study will increase our molecular understanding of B. minax olfaction, facilitating the development of OBP-based behavioral interference that is potentially useful for the integrated management of B. minax.

The monarch butterfly, Danaus plexippus L., has evolved to be insensitive to milkweed cardenolides via genetic modifications of Na+/K+-ATPase. There is concern for insecticide exposures near agriculture, with little information on monarch caterpillar toxicology. It is unclear how cardenolide insensitivity may affect the sensitivity of monarch caterpillars to pyrethroid insecticides. Additionally, potassium fertilizers may affect monarch caterpillar physiology and cardenolide sequestration. Here, we investigated the growth, survival, and development of caterpillars exposed to the cardenolide ouabain, bifenthrin, and potassium chloride (KCl) alone and in combination. Caterpillars were either exposed to 1) ouabain from third- to fifth-instar stage, 2) KCl at fifth-instar stage, 3) KCl and bifenthrin at fifth-instar stage, or 4) combinations of ouabain at third-instar stage + KCl + bifenthrin at fifth-instar stage. Caterpillar weight, diet consumption, frass, and survival were recorded for the duration of the experiments. It was observed that 1–3 mg ouabain/g diet increased body weight and diet consumption, whereas 50 mg KCl/g diet decreased body weight and diet consumption. Caterpillars feeding on KCl and treated with 0.2 µg/µl bifenthrin consumed significantly less diet compared to individuals provided untreated diet. However, there was no effect on survival or body weight. Combinations of KCl + ouabain did not significantly affect caterpillar survival or body weight following treatment with 0.1 µg/µl bifenthrin. At the concentrations tested, there were no effects observed for bifenthrin sensitivity with increasing cardenolide or KCl concentrations. Further studies are warranted to understand how milkweed-specific cardenolides, at increasing concentrations, and agrochemical inputs can affect monarch caterpillar physiology near agricultural landscapes.

Synthetic insecticide application is one tactic for reducing boll weevil, Anthonomus grandis grandis Boheman (Coleoptera: Curculionidae), infestations during the cotton, Gossypium hirsutum L., reproductive stage. We assessed the susceptibility of the boll weevil and its natural enemies to ethiprole (mode of action 2B), a phenylpyrazole insecticide, and diagnostic concentrations of ethiprole indicative of boll weevil susceptibility. Differences in the lethal concentrations of ethiprole were calculated with susceptibility ratios based on LC₅₀ ranging from 2.89- to 10.34-fold relative to a natural susceptible population.The lowest and the highest recommended field rates of ethiprole, 100 and 200 g a.i./ha, produced residues that caused 83.3% and 93.7% mortality of weevils caged with cotton leaves from field-treated plants for 8 d. We found that ethiprole was less toxic than fipronil to the boll weevil parasitoid Bracon vulgaris Ashmead (Hymenoptera: Braconidae) and to the red imported fire ant, Solenopsis invicta Buren (Hymenoptera: Formicidae), while fipronil was highly toxic to both. Adult earwigs, Euborellia annulipes Lucas (Dermaptera: Anisolabididae), were relatively tolerant to ethiprole and fipronil at the highest field rates. Pooled LC₅₀-and LC₉₅-concentrations of ethiprole calculated from studied populations were used as diagnostic for boll weevil mortality, and the outcome fitted to the expected mortality for boll weevil populations from different locations serving for further control failure assessment. Ethiprole appears to be suitable for boll weevil control with low impact on natural enemy communities.

Western corn rootworm, Diabrotica virgifera virgifera LeConte, biology is tied to the continuous availability of its host (corn, Zea mays L.). Annual rotation of corn with a nonhost, like soybean (Glycine max (L.) Merrill) was a reliable tactic to manage western corn rootworm. Behavioral resistance to annual crop rotation (rotation resistance) allowed some eastern U.S. Corn Belt populations to circumvent rotation by laying eggs in soybean and in cornfields. When active in soybean, rotation-resistant adults commonly consume foliage, in spite of detrimental effects on beetle survival. Rotation-resistant beetle activity in soybean is enabled by the expression of certain proteinases and an adapted gut microbiota that provide limited protection from soybean antiherbivore defenses. We investigated the effects of corn and soybean herbivory on rotation-resistant female survival and initiation of flight using mortality assays and wind tunnel flight tests. Among field-collected females tested with mortality assays, beetles from collection sites in a cornfield survived longer than those from collection sites in a soybean field. However, reduced survival due to soybean herbivory could be restored by consuming corn tissues. Field-collected beetles that fed on a soybean tissue laboratory diet or only water were more likely to fly in a wind tunnel than corn-feeding beetles. Regardless of collection site and laboratory diet, 90.5% of beetles that flew oriented their flights upwind. Diet-related changes in the probability of flight provide a proximate mechanism for interfield movement that facilitates restorative feeding and the survival of females previously engaged in soybean herbivory.

Rotation-resistant western corn rootworm females feeding on soybean tissues experience reduced survival in mortality assays and display increased flight probability (which may facilitate flight back to a cornfield where consumption of host tissues improves survival potential and facilitates maturation of eggs). The consequences of soybean herbivory provide a proximal mechanism for behavioral resistance to crop rotation. Increased egg-laying probability while feeding on soybean tissues, facilitation of egg maturation while feeding on corn tissues, and interfield movement are previously documented consequences.

Efficient separation of insects from plant material for quantification and collection is an important component of entomological research. This paper reports on a novel, easily replicable container designed to efficiently collect two different biological control agents dispersing from hemlock (Tsuga spp.) foliage infested with the invasive hemlock woolly adelgid (HWA), Adelges tsugae Annand (Hemiptera: Adelgidae). The container utilizes a simplified Berlese-style funnel design to collect Laricobius spp. (Coleoptera: Derodontidae) larvae dropping from the foliage into a removable bottom jar, a central jar to house the foliage sample, and a removable top jar to collect adult silver flies (Leucopis spp., Diptera: Chamaemyiidae) emerging from puparia on the twigs. The efficacy of two designs (with and without a funnel leading to the top collection jar) was evaluated using western hemlock [Tsuga heterophylla (Raf.) Sarg.] foliage naturally colonized with HWA and the two predator genera. All Laricobius larvae were effectively collected in the bottom jar, and the addition of an inverted funnel leading to the top collection jar increased the proportion of Leucopis flies reaching the target jar from 60% to 94%. This ‘Lari-Leuco’ container is presented as a research and motoring tool to benefit the integrated pest management program for HWA in eastern North America and for potential use in simultaneously separating ascending and descending life stages in other insect-plant or predator-prey systems.

The red palm weevil (RPW), Rhynchophorus ferrugineus (Oliver) is an important pest of palms that causes significant damage by boring into and feeding within palm stem tissues. Here, we studied the proteolytic process of Cry3Aa in the RPW to understand the mechanism of Cry toxicity. The bioassays showed that Cry3Aa toxin is weakly toxic to the RPW. Proteolytic activation assays indicated the Cry3Aa protein is digested into smaller fragments than the 55-kDa activated fragments under different conditions. In particular, at higher mass ratios of gut protease and Cry3Aa protein (5:1, 2:1, and 1:1, respectively), and at 36.9°C for 16 h in a solution of pH 8.6, the Cry3Aa protoxin is over-digested by the gut proteases of weevil larvae. Moreover, the zymogram analysis of the gut proteases revealed the RPW larvae harbors intestinal digestive enzymes mainly composed of serine proteases.This study describes the proteolytic activation process of Cry3Aa in the midgut of RPW larvae.

The larval-pupal endoparasitoid Diachasmimorpha longicaudata (Ashmead, 1905) is considered one of the main biological control agents of fruit flies Ceratitis capitata Wiedemann (1824) and Anastrepha fraterculus Wiedemann (1830) in Central and South America. The application of pesticides for disease and insect-pest management in fruit species may have adverse effects on the parasitoid. The objective of this study was to evaluate the lethal and sublethal effects via residual contact of commercial pesticide formulations on D. longicaudata. The active ingredients thiamethoxam, indoxacarb, chlorpyrifos, deltamethrin, spinetoram, spinosad, phosmet, lambda-cyhalothrin, malathion, dimethoate, and methidationa showed high toxicity to adults (100% mortality) after 96 h and were classified as harmful (Class 4). In contrast, the formulations of azadirachtin (Agroneem 850 EC, Azact 2.4 EC, Azamax 12 EC, and Fitoneem 850 EC), chlorantraniliprole, bordeaux mixture, sulfur, lufenuron, lime sulphur, novalurom, and mancozeb were rated as innocuous (<10% mortality). In addition, the formulations azadirachtin did not reduce the parasitism and the emergence rate of the F₀ generation, the same pesticides added to chlorantraniliprole, azadirachtin A+B (Agroneem 850 EC), and lufenuron did not cause reduction in parasitism and emergence rate of the F₁ generation of D. longicaudata. The use of pyrethroids, organophosphates, spinosyns, oxadiazines, and neonicotinoids should be used with caution in IPM programs. While pesticides chlorantraniliprole, azadirachtin formulations, bordeaux mixture, lufenuron, lime sulphur, and mancozeb do not cause lethal and sublethal effects for D. longicaudata adults. The results of this study provide important information for use in integrated pest management programs for fruit fly management.

A warmer climate and the development of pesticide resistance have led to an increase in the number of generations and higher population densities of California red scale, Aonidella aurantii (Maskell) in central California. Commercial citrus orchard studies were conducted to determine how effective mating disruption using CheckMate CRS was at reducing A. aurantii. In 2016–2017, two replicated trials were conducted in 15.0–15.5 ha orchards and in 2018 and 2019, one half of each of 10 orchards 8–16.2 ha in size were treated with CheckMate CRS. Mating disruption significantly suppressed male captures for up to 8 mo. Average reductions of 90% in cumulative male flight trap catches were recorded, twig and leaf infestations were reduced by 95%, and highly scale-infested fruit were reduced by 75% in the CheckMate CRS plots for the 2018 and 2019 trials. In seven of 12 sites the percentage of highly infested fruit was reduced below 0.5%. Leaf and twig infestations in August and reductions in male captures during the 4th flight were strongly related to the percentage of highly infested fruit at the end of the season and could be used as predictors of the success of mating disruption. The results of the study indicated mating disruption using CheckMate CRS can be an effective method to reduce California red scale populations throughout the 4+ generations that occur in central California. Mating disruption has the potential to reduce or eliminate pesticide applications, especially in low scale density situations.

Grapholita molesta is one of the most important fruit pests worldwide. Abamectin is a biological pesticide frequently used to control fruit borers like G. molesta in part owing to its translaminar properties. In this study, we characterized the toxicity of abamectin to G. molesta larvae using the diet incorporation method. The sublethal effects of abamectin on the development, reproduction, detoxification enzyme activity, and related gene expression of G. molesta were assessed. The results showed that the LC₂₀ and LC₅₀ values of the insecticide against G. molesta 72 h post-treatment were 1.17 mg L^–1 and 5.85 mg L^–1, whereas the LC₂₀ and LC₅₀ values 96 h post-treatment were 0.34 mg L^–1 and 3.63 mg L^–1. When compared to the control, sublethal concentrations of abamectin 1) significantly increased the mortality of the larvae, prepupae, and pupae of G. molesta, 2) prolonged the duration of 3rd to 5th instar larva, prepupal and pupal periods, 3) shortened the longevity of adults, and 4) reduced female fecundity. The enzymatic activity of glutathione S-transferase (GST) varied significantly after exposure to sublethal concentrations of abamectin, but the cytochrome P450 monooxygenases and carboxylesterase activity were not significantly affected. Thirteen of the 25 GST genes were significantly upregulated under different sublethal concentrations of abamectin. The combined findings increase our understanding of the effects of abamectin on G. molesta and the potential role of GSTs in the metabolic interactions of abamectin in this pest, and have applications for more rational and effective use of abamectin to control G. molesta.

The mealybug, Planococcus ficus (Signoret), is a primary vineyard pest in California and other grape-growing regions throughout the World. Mating disruption programs are commercially available to manage Pl. ficus, but widespread adoption has been limited, in part, by high costs compared with insecticide programs. To improve mating disruption economic effectiveness, different deployment technologies (passive, aerosol, and microencapsulated formulations) were individually examined. Adult male Pl. ficus captures in pheromone traps and categorical ratings of vine infestation or crop damage suggest that all deployment strategies lowered mealybug densities or damage. Using passive dispensers, deployment rates of 310 and 465 per ha lowered Pl. ficus crop damage similar to 615 per ha, a rate commonly used in commercial operations; reduced rates would lower product and deployment costs. Meso dispensers, containing more a.i., deployed at 35 per ha did not have a treatment impact, but a microencapsulated formulation and aerosol canisters lowered male flight captures and/or crop damage. Male mealybug flight activity was greatest from 0500–1100 hr, which coincided with temperatures >16° and <32°C.These restricted times and temperatures suggest programable dispensers might allow pheromone deployment to coincide only with flight patterns. A large field trial using passive dispensers found greater treatment separation after 3 yr of pheromone deployment. Discrepancies in results among vineyards may be related to Pl. ficus density, but combined results from all trials suggest that different deployment technologies can be used to impact Pl. ficus densities and damage, even at reduced rates, especially with continued use over multiple seasons.

Several species of drywood termites, subterranean termites, and fungus-growing termites cause extensive economic losses annually worldwide. Because no universal method is available for controlling all termites, correct species identification is crucial for termite management. Despite deep neural network technologies' promising performance in pest recognition, a method for automatic termite recognition remains lacking. To develop an automated deep learning classifier for termite image recognition suitable for mobile applications, we used smartphones to acquire 18,000 original images each of four termite pest species: Kalotermitidae: Cryptotermes domesticus (Haviland); Rhinotermitidae: Coptotermes formosanus Shiraki and Reticulitermes flaviceps (Oshima); and Termitidae: Odontotermes formosanus (Shiraki). Each original image included multiple individuals, and we applied five image segmentation techniques for capturing individual termites. We used 24,000 individual-termite images (4 species × 2 castes × 3 groups × 1,000 images) for model development and testing. We implemented a termite classification system by using a deep learning–based model, MobileNetV2. Our models achieved high accuracy scores of 0.947, 0.946, and 0.929 for identifying soldiers, workers, and both castes, respectively, which is not significantly different from human expert performance. We further applied image augmentation techniques, including geometrical transformations and intensity transformations, to individual-termite images. The results revealed that the same classification accuracy can be achieved by using 1,000 augmented images derived from only 200 individual-termite images, thus facilitating further model development on the basis of many fewer original images. Our image-based identification system can enable the selection of termite control tools for pest management professionals or homeowners.

Strategic responses to invasive Latrodectus widow spiders are a global challenge due to the risks they pose to health and ecosystems. Chemical strategies involving the use of pyrethroids are effective against adult spiders, but as their populations rebound, chemical control becomes costly and unsustainable for eradication. A major obstacle is the inefficacy of insecticides against eggs, which are covered by a protective silk egg sac. Eradication of invasive spiders must focus on destroying progeny. Here, the responses of eggs in egg sacs of two invasive Latrodectus spiders in Japan (Latrodectus hasseltii (Thorell) and Latrodectus geometricus (C.L. Koch)) to short-term dry-heat exposure were examined. To test whether the dry-heat tolerance of the egg sacs of both spider species differed, lethal temperature (LT) was determined based on the hatching rate of eggs from egg sacs subjected to a range of temperatures. Hatching in both species failed completely when the egg sacs were exposed to temperatures of 55°C and above for 10 min, but the LT to reduce hatching by 90% (LT₉₀) differed significantly between L. hasseltii (50. 9°C) and L. geometricus (52. 8°C). Our study highlights the efficacy of dry heat in suppressing hatching and thus shows the possibility for effective extermination of these noxious invasive pests. Further exploration and investigation of the effects of humidity and heat exposure time on egg sacs under field conditions are needed to guide Latrodectus spider control strategies.

Subterranean termite control methods using chitin synthesis inhibitors (CSIs) aim at eliminating colonies that feed upon a bait formulation. Several benzoylurea active ingredient formulations are currently commercially available as alternative termite management strategies to liquid termiticides. Individual workers need to molt on a regular basis and CSIs interfere with such molting process, allowing sufficient time for the acquisition of a colony-wide lethal dose prior to widespread mortality. As workers progressively die, the colony eventually collapses, leaving only soldiers and primary reproductives that starve to death. One common observation is that young workers often die early owing to their relatively short molting cycle. However, the absence of brood in dying colonies raises questions about the potential fate of eggs laid by the queen. This study aims to determine if CSI baits also terminate the ability of a colony to produce a new cohort of workers by disabling the ongoing brood development. Incipient termite colonies were used to test the impact of noviflumuron on the queen's ability to lay eggs and on the eggs' ability to hatch. Our results showed that queens in colonies exposed to CSI not only initially laid less eggs than the control queens, but eggs also did not develop and were progressively cannibalized, eventually leading to colony establishment failure. This result implies that queens of mature colonies exposed to CSI would lose the ability to lay viable eggs as the colony collapses, leading to an absence of worker replacement, aiding in colony elimination.

The modern resurgence of the common (Cimex lectularius L.) and tropical bed bugs (C. hemipterus [F.]) is thought to be primarily due to insecticide resistance. While there are many reports on insecticide resistance mechanisms in C. lectularius, such information in C. hemipterus is limited. We examined dichloro-diphenyl-trichloroethane (DDT), malathion, deltamethrin, permethrin, lambda-cyhalothrin resistance, and the underlying mechanisms in several C. hemipterus strains (Australia: Queensland [QLD-AU]; Malaysia: Kuala Lumpur [KL-MY], Tanjung Tokong [TT-MY], Christian [CH-MY], and Green Lane [GL-MY]). We used a surface contact method, synergism studies (utilizing piperonyl butoxide [PBO], S,S,S-tributyl phosphorotrithioate [DEF], and diethyl maleate [DEM]), and molecular detection of kdr mutations. Results demonstrated that all C. hemipterus strains possessed high resistance to DDT and the pyrethroids and moderate to high resistance to malathion. Synergism studies showed that deltamethrin resistance in all strains was significantly (P < 0.05) inhibited by PBO. In contrast, deltamethrin resistance was not affected in DEF or DEM. Similar findings were found with lambda-cyhalothrin resistance. Malathion resistance was significantly (P < 0.05) reduced by DEF in all strains. Resistance to DDT was not affected by DEM in all strains. Multiple kdr mutations (M918I, D953G, and L1014F) were detected by molecular analyses. TT-MY strain was found with individuals possessing three kdr mutation combinations; D953G + L1014F (homozygous susceptible: M918), M918I + D953G + L1014F (heterozygous resistant: I918), and M918I + D953G + L1014F (homozygous resistant: I918). Individuals with M918I + D953G + L1014F (homozygous resistant: I918) survived longer on deltamethrin (>12 h) than those (≤1 h) with other combinations. M918I + L1014F mutations most likely conferred super-kdr characteristic toward pyrethroids and DDT in C. hemipterus.

We previously extracted and purified a chromene amide from Amyris texana and found this scaffold is moderately insecticidal and thus, this study aimed to test the insecticidal properties of 13 synthetically derived chromene analogs to the fall armyworm (FAW, Spodoptera frugiperda). Microinjection of chromenes with alcohol or aldehydes substitutions at the meta position on the benzopyran moiety led to moderate toxicity that was approximately 2- to 3-fold less toxic when compared to permethrin, yet microinjection of differently substituted chromenes exhibited little to no toxicity. Similarly, chromenes with alcohol or aldehydes substitutions at the meta position on the benzopyran moiety were among the most toxic chromenes studied through ingested exposure. In addition to acute toxicity, select chromenes significantly increased the percentage of developmental defects upon eclosion that prevented adult moths from being capable of flight, suggesting these compounds alter development. Interestingly, microinjection yielded differing signs of intoxication between alcohol and aldehyde substitutions where the alcohol resulted in flaccid paralysis and lethargy whereas aldehyde led to tonic contractions and hyperactivity. These contrasting signs of intoxication were also observed in electrophysiological assays where alcohol substitutions led to the depression of central neuron firing activity and aldehyde substitutions led to hyperexcitation of central neurons. In summary, the chromene amides led to acute lethality and/or altered developmental trajectories of FAW, yet the high doses required for acute mortality suggest these scaffolds hold relatively little promise for development into FAW-directed insecticides but may represent novel growth regulators for FAW.

Widespread field-evolved resistance of bollworm [Helicoverpa zea (Boddie)] to Cry1 and Cry2 Bt proteins has threatened the utility of Bt cotton for managing bollworm. Consequently, foliar insecticide applications have been widely adopted to provide necessary additional control. Field experiments were conducted across the Mid-South and inTexas to devise economic thresholds for foliar insecticide applications targeting bollworm in cotton. Bt cotton technologies including TwinLink (TL; Cry1Ab+Cry2Ae), TwinLink Plus (TLP; Cry1Ab+Cry2Ae+Vip3Aa), Bollgard II (BG2; Cry1Ac+Cry2Ab), Bollgard 3 (BG3; Cry1Ac+Cry2Ab+Vip3Aa), WideStrike (WS; Cry1Ac+Cry1F), WideStrike 3 (WS3; Cry1Ac+Cry1F+Vip3Aa), and a non-Bt (NBT) variety were evaluated. Gain threshold, economic injury level, and economic thresholds were determined. A 6% fruiting form injury threshold was selected and compared with preventive treatments utilizing chlorantraniliprole. Additionally, the differences in yield from spraying bollworms was compared among Bt cotton technologies. The 6% fruiting form injury threshold resulted in a 25 and 75% reduction in insecticide applications relative to preventive sprays for WS and BG2, respectively. All Bt technologies tested in the current study exhibited a positive increase in yield from insecticide application. The frequency of yield increase from spraying WS was comparable to that of NBT. Significant yield increases due to insecticide application occurred less frequently in triple-gene Bt cotton. However, their frequencies were close to the dual-gene Bt cotton, except for WS. The results of our study suggest that 6% fruiting form injury is a viable threshold, and incorporating a vetted economic threshold into an Integrated Pest Management program targeting bollworm should improve the sustainability of cotton production.

The beet armyworm, Spodoptera exigua (Hübner), is a major insect pest in shallot in Java, Indonesia. To control this insect, farmers rely on the use of a narrow spectrum of insecticides leading to resistance development. The objective of this study was to determine the level of resistance of S. exigua populations from different areas of Java to the most widely used insecticides, chlorfenapyr, methomyl, and emamectin benzoate, as well as to cyantraniliprole which was recently introduced in Indonesia. Spodoptera exigua populations were collected from three major shallot production areas in Java, the Districts of Brebes, Nganjuk, and Bantul. Based on the LC₅₀ values, resistance ratios of S. exigua populations compared to the most susceptible population from the sub-district Pundong in Bantul varied from very low to very high: 1.1- to 113.8-fold for chlorfenapyr, 16.4- to 223.6-fold for methomyl, and 1.5- to 51.7-fold for emamectin benzoate. Spodoptera exigua populations from the same location were more susceptible to cyantraniliprole, with a resistance ratio of 4.0- to 12.1-fold. These findings indicate that the field populations of S. exigua collected from major shallot areas in Java were resistant to chlorfenapyr and methomyl and less resistant to emamectin benzoate, whereas they were still susceptible to cyantraniliprole. The populations from Brebes and Nganjuk were more resistant to chlorfenapyr, methomyl, and emamectin benzoate than those collected from Bantul. To prevent the further increase of resistant of S. exigua populations, IPM principles need to be applied including the alternate use of insecticides with different modes of action.

Alternative splicing of protein coding genes plays a profound role in phenotypic variation for many eukaryotic organisms. The development of high-throughput sequencing and bioinformatics algorithms provides the possibility of genome-wide identification of alternative splicing events in eukaryotes. However, for the brown planthopper Nilaparvata lugens, a destructive pest of rice crops, whole-genome distribution of alternative splicing events and the role of alternative splicing in the phenotypic plasticity of virulence have not previously been estimated. Here, we developed an analysis pipeline to identify alternative splicing events in the genome of N. lugens. Differential expression analysis and functional annotation were performed on datasets related to different virulence phenotypes. In total, 27,880 alternative splicing events corresponding to 9,787 multi-exon genes were detected in N. lugens. Among them, specifically expressed alternative splicing transcripts in the virulent Mudgo population were enriched in metabolic process categories, while transcripts in the avirulent TN1 population were enriched in regulator activity categories. In addition, genes encoding odorant receptor, secreted saliva protein and xenobiotic metabolic P450 monooxygenase showed different splicing patterns between Mudgo population andTN1 population. Host change experiment also revealed that an isoform of a P450 gene could be specially induced by the stimulation of resistant rice variety Mudgo. This research pioneered a genome-wide study of alternative gene splicing in the rice brown planthopper. Differences in alternative splicing between virulent and avirulent populations indicated that alternative splicing might play an important role in the formation of virulence phenotypes in N. lugens.

Australia is the third largest exporting country of cereals and a leader in other major commodity crops, yet little data exist on pesticide usage patterns in agriculture. This knowledge gap limits the management of off-target chemical impacts, such as the evolution of pesticide resistance. Here, for the first time, we quantify spatial patterns in neonicotinoid applications in Australia by coalescing land use data with sales and market research data contributed by agrichemical and agribusiness companies. An example application to resistance management is explored through the development of recommendations for the cosmopolitan crop pest, Myzus persicae (Sulzer) (Hemiptera: Aphididae), utilizing spatial statistical models. This novel dataset identified Australian neonicotinoid usage patterns, with most neonicotinoid products in Australia applied as cereal, canola, cotton and legume seed treatments and soil applications in sugarcane. Importantly, there were strong regional differences in pesticide applications, which will require regionally specific strategies to manage off-target impacts. Indeed, the estimated spatial grid of neonicotinoid usage demonstrated a statistically significant influence on the distribution of M. persicae neonicotinoid resistance, indicating off-target impacts are unevenly distributed in space. Future research on neonicotinoid usage will be supported by the spatial grids generated and made available through this study. Overall, neonicotinoid pesticides are widely relied upon throughout Australia's plant production systems but will face increasing pressure from resistance evolution, emerging research on off-target impacts, and stricter regulatory pressures.

Volatile organic compounds from various plants have received popular interest as one of the vector control tools due to their eco-friendliness and insect-repellent activities. In this study, an excito-repellency assay system was used to examine the noncontact repellency, contact excitation, and knockdown (KD) effects of guava leaf (Psidium guajava L.) oil against Anopheles minimus (Theobald), Anopheles epiroticus (Linton & Harbach), and Culex quinquefasciatus (Say). The organic components of guava oil were identified by gas chromatography–mass spectrometry analysis with DL-limonene (17.4%), cymene (5.49%), and α-terpinene (5.20%) as the major constituents. At concentrations of 2.5 and 5.0%, 100% escape of An. minimus was recorded in the contact assay and 96–98% escape in the noncontact assay. Guava oil stimulated potent irritant (92% escape) and repellent (61–86% escape) effects against Cx. quinquefasciatus. A lower repellency action was observed against An. epiroticus (17–20% escape). No KD effect was observed for guava oil against An. minimus and Cx. quinquefasciatus at any concentration. However, An. epiroticus was more prone to KD effects, with the highest percentage KD (44% in nonescape group) observed with 5.0% guava oil in the noncontact assay. Mortalities of 35% and 11% were observed for An. epiroticus in the nonescape groups in the contact and noncontact assays, respectively. Concentrations of 1.0% and 2.5% guava oil led to <2% mortality in An. minimus. Our findings highlight guava oil as a promising plant-based mosquito repellent that can be included in insecticide formulations for future mosquito control programs.

Eotetranychus kankitus is an important mite pest in citrus, but molecular data on the developmental processes of E. kankitus are lacking. The different development stages mix of E. kankitus was used to sequence for transcriptome and small RNAs to identify genes and predict miRNAs associated with sesquiterpenoid and ecdysteroid biosynthesis and signaling pathways. More than 36 million clean reads were assembled and 67,927 unigenes were generated. Of the unigenes, 19,300 were successfully annotated through annotation databases NR, SwissProt, COG, GO, KEGG, PFAM, and KOG. The transcripts were involved in sesquiterpenoid biosynthesis (11 genes) and ecdysteroid biosynthesis and signaling pathway (13 genes). Another, small RNA library was obtained and 31 conserved miRNAs were identified. Five most abundant miRNAs were Ek-miR-5735, Ek-miR-1, Ek-miR-263a, Ek-miR-184, and Ek-miR-8.The target genes related to sesquiterpenoid and ecdysteroid showed that 10 of the conserved miRNAs could potentially target the sesquiterpenoid and ecdysteroid pathway according to four-prediction software, sRNAT, miRanda, RNAhybrid, and Risearch2. Thus, the results of this study will provide bioinformatics information for further molecular studies of E. kankitus which may facilitate improved pest control strategies.

Uridine diphosphate glucosyltransferases (UGTs), which are phase II detoxification enzymes, are found in various organisms.These enzymes play an important role in the detoxification mechanisms of plant allelopathy and in insects. Artemisia vulgaris L. (Asterales: Asteraceae: Artemisia) essential oil has strong contact toxicity to Tribolium castaneum Herbst (Coleoptera: Tenebrionidae) larvae. However, the effect of A. vulgaris essential oil on UGTs is unclear. In this study, A. vulgaris essential oil was shown to significantly induce the expression of the TcUgt86Dg transcript. Furthermore, treatment of TcUgt86Dg-silenced individuals with A. vulgaris essential oil resulted in higher mortality than for the control individuals, indicating that TcUgt86Dg is involved in detoxification of A. vulgaris essential oil in T. castaneum. The developmental expression profile showed that the expression of TcUgt86Dg in late adults was higher than in other developmental stages. Furthermore, the expression profile in adult tissues revealed higher expression of TcUgt86Dg in the head, antenna, fat body, and accessory gland than in other tissues. These data show that TcUgt86Dg may be involved in the metabolism of exogenous toxins by T. castaneum; thus, our results have elucidated one possible mechanism of resistance to A. vulgaris essential oil and provide a theoretical basis for a control scheme for T. castaneum.

Vitellogenin (Vg) and the target of rapamycin (TOR) are important genes involved in insect reproduction regulation. In this work, the full lengths of the Vg (SfVg) and TOR (SfTOR) genes of the white-backed planthopper Sogatella furcifera were cloned. The expression pattern in females showed that SfVg was highly expressed in fat bodies, and SfTOR was highly expressed in Malpighian tubules. After silencing SfVg or SfTOR, female adults did not deposit eggs. Their ovarian development was delayed, and yolk protein deposition in the oocytes was reduced. However, wild-type females mated with SfTOR-silenced males could lay eggs and produce offsprings normally. The dissections of testes and accessory glands of males with SfTOR knockdown showed that their development was not affected. Therefore, the silencing of the SfVg or SfTOR genes can effectively inhibit female reproduction, but SfTOR knockdown has no significant effect on male reproductive capacity. Furthermore, silencing SfTOR can cause SfVg expression to decrease significantly. All of the above results revealed that SfVg and SfTOR are essential for white-backed planthopper reproduction and may provide a potential target for pest control.

Injury by herbivores is a major biotic stress that limits soybean [Glycine max (L.) Merrill] crop production. Among the main soybean insect pests, Anticarsia gemmatalis Hübner is responsible for causing significant economic damage in soybean. The primary management strategy for this insect is chemical control and use of Bt transgenic soybean. Alternative strategies, such as host plant resistance, are considered an efficient and less-aggressive method, especially in association with other strategies as part of an integrated pest management (IPM) approach. In this study, we evaluated 30 soybean genotypes to verify antixenosis expression through oviposition, attractiveness, and food consumption tests. From this, we selected 13 promising genotypes to verify the possible presence of antibiosis. Our results suggest that antixenosis was found in genotypes ‘TMG 133' RR, ‘TMG 1179' RR, ‘IAC 19’, ‘IAC 17’, ‘IAC 100’, D75-10169, and IAC 78-2318. By influence on behavior and negative impact on larval viability, antixenosis and antibiosis were indicated for the genotypes IAC 74-2832, ‘IAC 19’, ‘IAC 17’, ‘IAC 100’, and PI 274454. ‘TMG 7062’ IPRO was found to provide antibiosis resistance by negatively affecting larval development and viability. Because of reduced food consumption by larvae, antixenosis was indicated for ‘IAC 24’. These genotypes should be considered in soybean breeding programs focusing on soybean resistance to A. gemmatalis.

Fully trapped survey designs are widely used to delimit adventive pests populations that can be detected using traps and lures. Delimitation includes verifying the presence of the pest and determining its spatial extent. The size and shape of the survey design and the density of traps can vary; however, resulting variation in detecting efficiency is often unknown. We used a trapping network simulation model with diffusion-based insect movement to investigate delimiting survey trapping design performance for fully trapped and some modified designs. Simulations included randomized outbreak locations in a core area and a duration of 30 d. We assessed impacts of insect dispersal ability, grid size and shape, and trap attractiveness and density on survey performance, measured as mean probability of capturing individual pests [p(capture)]. Most published grids are square, but circles performed equally well and are more efficient. Over different grid sizes, p(capture) increased for insects with greater dispersal ability but was generally unresponsive to size because most captures occurred in central areas. For low dispersing insects, the likelihood of egress was approximately zero with a 3.2-km square grid, whereas an 11.3-km grid was needed to contain highly vagile insects. Trap attractiveness affected p(capture) more strongly than density: lower densities of poorly attractive traps may underperform expectations. Variable density designs demonstrated potential for cost savings but highlighted that resource-intensive outer bands are critical to boundary determination. Results suggesting that many grids are oversized need empirical verification, whereas other principles, such as using circular shapes, are readily adoptable now.

In the present work, a series of tests were performed to examine the effect of previously captured adults in floor traps, on the captures of Trogoderma granarium Everts and T. variabile Ballion, in single species releases. In a first series of tests, a single trap with kairomone oil and dead adults of single or both species was placed in a plastic container, which was used as the release arena. Subsequently, adults of both species were separately released in the arena, and the adult capture was recorded 24 h later. In a second series of trials, two traps were placed in the same arena, containing different numbers of prior seeded adults, and adult capture was similarly recorded. For T. granarium, in all cases, more adults were found inside the trap than under the trap or on the trap walls. In contrast, for T. variabile, most adults were recorded on the trap walls. In general, T. variabile had a stronger response to the traps than T. granarium.This work illustrates that previous captures of either species do not negatively affect captures of new live adults and may tend to enhance captures in some cases. This has important implications for the use of traps for the biosureveillance of these two important dermestid species.

Preventing insect infestations is a critical component for establishing a pest management plan for stored-product insects. Long-lasting insecticide-treated netting (LLIN) is a potential tool to reduce insect movement by providing a chemical barrier, where insects may be able to physically pass through but ultimately die after exposure to the netting. Sublethal effects, such as reduced movement immediately after exposure and reduced ability to colonize, have been reported. Here we examine the sublethal effects of exposure to LLIN on two beetle species, Trogoderma variabile Ballion, warehouse beetle, and Tribolium castaneum Herbst, red flour beetle. We found that both female and male T. castaneum exposed to LLIN produced significantly less adult progeny than those exposed to untreated netting. Adult progeny output did not differ for T. variabile, but survivorship increased in T. variabile females exposed to LLIN. Importantly, the overall net reproductive rate was significantly decreased for both T. variabile and T. castaneum. The number of copulation attempts did not differ between males or females exposed to LLIN compared to untreated netting, but males exposed to LLIN showed increased durations of attempted and successful copulation events. This research demonstrates that the implications of LLIN exposure extend past direct mortality, with sublethal effects on reproductive output potentially increasing the effectiveness of this tool for preventing insect infestations.
