In general, African catfish shows higher survival rates in the dark conditions than in the light conditions. In this study, larval behavior of African catfish was observed under 0, 0.01, 0.1, 1, 10, and 100 lx using a CCD camera to investigate the reason why African catfish larvae show higher survival rates in dark conditions. The larvae showed significantly higher swimming activity under 0, 0.01, and 0.1 lx than that under 10 and 100 lx. The larvae also showed significantly increased aggressive behavior under 10 and 100 lx; the swimming larvae attacked resting individuals more frequently under 10 and 100 lx than under 0, 0.01, and 0.1 lx. The aggressive behavior and sharp teeth of the attacking larvae appeared to induce skin surface lesions on injured larvae. Chemical substances were then generated from the injured skin surface, and these chemical stimuli triggered cannibalistic behavior in other fish near the injured fish. The results of this study demonstrate that the higher survival rates of African catfish larvae under dark conditions are a result of inactivity and subsequent increase in chemical releasing stimuli concentrations around inactive individuals that triggers feeding behavior in nearby active catfish. Therefore, we recommend larval rearing of African catfish in dark or dim conditions, as it improves catfish survival rates.
Olfaction-based reproductive isolation is widely observed in animals, but little is known about the genetic basis of such isolation mechanisms. Two species of sibling amphibious sea snakes, Laticauda colubrina and L. frontalis live in Vanuatu sympatrically and syntopically, but no natural hybrids have been reported. Adult females of both taxa possess distinctive lipids in the skin, and male L. frontalis distinguishes conspecific females based on olfactory cues. To shed light on the molecular basis of the evolution of olfaction-based isolation mechanisms, olfactory receptor (OR) gene repertoires of both taxa were identified using pyrosequencing-based technology, and orthologous OR gene sets were identified. Few species-specific gene duplications or species-specific gene losses were found. However, the nonsynonymous-to-synonymous substitution rate ratio was relatively higher between orthologous OR genes of L. frontalis and L. colubrina, indicating that L. frontalis and L. colubrina have evolved to possess different olfactory senses. We suggest that L. frontalis and L. colubrina have evolved allopatrically, and this may be a byproduct of the allopatric evolution, and that this dissimilarity may function as a premating isolation barrier, since L. frontalis has returned to the ancestral range (Vanuatu).
The Japanese crested ibis Nipponia nippon is a critically threatened bird. We assessed genetic diversity and structure in the Sado captive population of the Japanese crested ibis based on 24 and 50 microsatellite markers developed respectively for the same and related species. Of a total of 74 loci, 19 showed polymorphisms in the five founder birds of the population, and therefore were useful for the analysis of genetic diversity and structure. Genetic diversity measures, A, ne, He, Hoand PIC, obtained by genotyping of the 138 descendants were similar to those of other species with population bottlenecks, and thus considerably low. The low level of genetic diversity resulting from such bottlenecks was consistent with the results of lower genetic diversity measures for the Sado captive relative to the Chinese population that is the source population for the Sado group as determined using previously reported data and heterozygosity excess by Hardy-Weinberg equilibrium tests. Further, individual clustering based on the allele-sharing distance and Bayesian model-based clustering revealed that the founder genomes were equally at population in total, and with various admixture patterns at individual levels inherited by the descendants. The clustering results, together with the result of inheritance of all alleles of the microsatellites from the founders to descendants, suggest that planned mating in captive-breeding programs for the population has succeeded in maintaining genetic diversity and minimizing kinship. In addition, the Bayesian model-based clustering assumed two different components of genomes in the Sado captive Japanese crested ibis, supporting a considerably low level of genetic diversity.
Melatonin and the plant hormone auxin (indole-3-acetic acid) have some structural similarity and, may thus exert comparable physiological effects on reproduction and growth. To test this possibility, I examined the effects of melatonin and auxin administration on reproductive and non-reproductive organ development in an animal model, the marsh rice rat Oryzomys palustris. Juvenile males housed under 14L:10D conditions were injected daily for four weeks with saline, melatonin, auxin, or melatonin and auxin, and the development of the testes and other organs was assessed. Melatonin alone significantly inhibited the development of the testes, seminal vesicles, Harderian glands, and overall somatic growth, but not the spleen. Auxin did not affect any endpoint measured. When melatonin was administered simultaneously with auxin, the melatonin effects dominated in suppressing reproduction and growth. The administration of melatonin or auxin in the drinking water produced results similar to the effects of melatonin and auxin injections reported herein. Lastly, both melatonin and auxin in the drinking water failed to alter any short photoperiod-induced reproductive inhibition. These data suggest that structural similarities between melatonin and auxin do not result in similar postnatal effects on reproductive and non-reproductive organ development on a long photoperiod and further suggest that melatonin and auxin do not operate through a common physiological mechanism.
Kisspeptins have emerged as potent regulators of the reproductive brain-pituitary-gonad (BPG) axis. Our previous study demonstrated that the brain of the chub mackerel (Scomber japonicus), a scombroid fish, expresses two kisspeptin-encoding genes, kiss1 and kiss2, and exhibits sexually dimorphic expression profiles. Recent studies strongly suggest that teleost Kiss1 and Kiss2 precursors produce mature Kiss1-pentadecapeptides (Kiss1–15) and Kiss2-dodecapeptides (Kiss2–12), respectively. In light of the above, the present study evaluated the potency of synthetic peptides of Kiss1–15, Kiss2–12, and a GnRH analog (GnRHa) on inducing gonadal development in sexually immature adult chub mackerel. Synthetic peptides were administered subcutaneously through mini-osmotic pumps. On day 45 post-administration, gonadosomatic index (GSI) values (%) of male fish treated with Kiss1–15 (1.82) significantly increased in comparison to initial control (0.33), final control (0.49), Kiss2–12 (0.24), and GnRHa (1.13)-treated fish. Interestingly, the testis of all Kiss1–15 treated fish revealed spermiation, and were full of spermatozoa. These fish showed significantly higher levels of pituitary fshβ and Ihβ mRNAs and circulating 11-ketotestosterone. GnRHa treated fish also revealed the presence of few spermatozoa in the testis. In females, no significant changes in GSI values were found between treatments; however, Kiss1–15- and GnRHa-treated fish showed prominent signs of vitellogenic onset, with many early yolk oocytes in their ovaries. Interestingly, Kiss1–15-treated fish exhibited higher levels of pituitary fshβ and circulating estradiol-17β. These results indicate that peripheral administration of Kiss1–15 and GnRHa can induce gonadal development in sexually immature chub mackerel.
Transcription activator-like effector nucleases (TALENs) are attractive and powerful molecular tools for targeted gene disruption because of their simple design and quick assembly. To evaluate the utility of TALENs in genome editing in Xenopus tropicalis, we prepared nine pairs of TALENs for the tyrosinase, noggin and MMP-9TH genes. All of the TALENs had some activity in a single-strand annealing assay using a cultured frog cell line, suggesting double-stranded DNA cleavage activity by the TALENs at their target site. The injection of mRNAs encoding TALENs into fertilized X. tropicalis embryos resulted in Cel-1 cleavage of the PCR fragment containing the target site amplified from embryo genomic DNA, indicating that a mutation in the target gene had occurred during embryogenesis. These mutations were confirmed by the sequencing of clones derived from the PCR fragments of genomic DNA. Patches of vitiligo were observed in tadpoles raised from fertilized eggs that had been injected with mRNAs of TALENs for the tyrosinase gene. TALENs containing the repeat variable di-residue (RVD) NN appeared to show more activity than TALENs containing RVD NK, although both RVD NN and NK preferentially associate with a G nucleotide.
We examined gill morphology and ventilatory function in the carpet shark family Parascylliidae using 14 preserved specimens of Parascyllium ferrugineum, P. variolatum, P. collare and Cirrhoscyllium japonicum, and two live specimens of P.ferrugineum and P.variolatum. Morphological examinations revealed eight morphological characteristics related to the fifth gill, based on comparisons with other elasmobranchs, viz. large fifth gill slit without gill filaments, anatomical modifications in the fourth ceratobranchial cartilage and coraco-branchialis muscle, and the hypaxialis muscle associated with the fifth gill arch. Ventilation examinations using dyed seawater and prey items showed different water flows through the gill slits for respiration and prey-capture actions. For respiration, water sucked into the mouth was expelled equally through the first to fourth gill slits via a “double-pump” action, there being no involvement of the fifth gill slit. In prey-capture, however, water sucked into the mouth was discharged only via the widely opened fifth gill slit. This form of water flow is similar to that in other benthic suction-feeding sharks (e.g., Chiloscyllium plagiosum), except for the active water discharge by wide expansion and contraction of the fifth parabranchial cavity. The latter is dependent upon the morphological modifications of the fourth and fifth gill arches, derived phylogenetically as a mechanistic suction specialization in Parascylliidae.
Otolith shape were investigated to identify two species of genus Dussumieria inhabiting the northern Oman Sea, south of Iran. The main aim of the investigation was to analyze otolith shape differences between these species. The sagittal otoliths of the rainbow sardine Dussumieria acuta Valenciennes, 1847, and the slender rainbow sardine Dussumieria elopsoides Bleeker, 1849, belonging to different length groups were described. The results showed two groups of special characteristics of the sagittal otoliths in D. acuta, and D. elopsoides. The first group is the characteristics that are useful to separate these species from other clupeid species, however these characteristics are closely related to one another in these species of Dussumieria genus. The second group comprises characteristics that vary due to genetically guided mechanisms and biological factors, but that may be useful to define species and are species-specific.
Gobiocypris rarus Ye et Fu (1983) is an endemic cyprinid fish in China, and is considered to be an endangered species. From a conservation perspective, its population structure is interesting. In the present study, morphological variation, including morphometric and meristic traits, was assessed among wild samples collected in the upper Yangtze River basin. There were no significant meristic differences between sexes or among populations, except for scales in lateral line (LS). However, there were significant morphometric differences not only between sexes but also among populations. In discriminant function analysis, the first four discriminant functions explained 75.5% and 78% of the among-population morphometric variation for males and females, respectively. Almost all the truss network morphometric traits significantly contributed to population discrimination. By using all of the truss network morphometric traits, overall random assignments of male and female individuals into their original population were both 73.5%. In addition, the degree of differentiation in phenotypic traits (PST) significantly exceeds that in neutral molecular markers (FST). However, no significant correlation between PST and FST was found in males or females. Overall, these results suggest that two evolutionary processes, including phenotypic plasticity and natural selection, may contribute to the morphological patterns observed in G.rarus.
In the Mongolian gerbil, bilateral common carotid artery occlusion (BCCAO) for several minutes induces ischemia and delayed neuronal cell death in the CA1 region of the hippocampus due to their incomplete Circle of Willis. In the present study, the expression of fibroblast growth factor 2 (FGF2), its receptors (FGFR1 and FGFR2), glial fibrillary acidic protein (GFAP), and isolectin B4 (ISLB4) was investigated by immunohistochemical and lectin-binding methods after BCCAO was performed for 5 min in gerbils. One day after BCCAO, the pyramidal cells of the CA1 region of the hippocampus showed degenerative changes and lowered expression of FGF2, FGFR1, and FGFR2. Three days after BCCAO, there was an increase in GFAP-positive astrocytes and ISLB4-positive microglial cells. From five to 10 days after BCCAO, intense neuronal cell death in the stria pyramidale of the hippocampal CA1 region was observed, as well as an increase in GFAP-positive astrocytes and decrease in ISLB4-positive microglial cells. These results indicate that transient forebrain ischemia induces neuronal cell death with lowered expression of FGF2 and its receptors, and that the activation of glial cells may not directly lead to neuronal cell death.
The phylogeny and related evolutionary history of rove beetles (Coleoptera, Staphylinidae) remain unclear. This study provides phylogenetic analyses for the family based on three genes (mitochondrial COI, nuclear protein-coding wingless and a portion of the ribosomal 28S rDNA) including 2413 bp for 104 taxa representing most major staphylinid lineages. The subfamilies Oxyporinae, Paederinae, Steninae, and Proteininae are all well-supported clades, as evidenced by all three inference methods, namely maximum parsimony, Bayesian inference, and maximum likelihood. From fossils available for calibration, the divergence time of the main lineages in the family is estimated based on an uncorrelated lognormal relaxed molecular clock analysis method. The molecular clock analysis suggests that the family Staphylinidae dates from approximately the Early Triassic epoch and the most lineages of the family started to radiate from the Late Jurassic to the Early Paleogene.
We present new data on the distribution, reproductive strategies, karyology, and taxonomic status of populations of freshwater planarians from Yemen. Nine populations were sampled and significant differences in their reproductive strategies and karyology are reported. The present study presents the first fully documented record of a naturally sexual, diploid (2n = 18) population of a Dugesia species in the eastern part of the Afrotropical region. Morphological characters combined with karyological data suggest that these Dugesia populations from Yemen represent a new species, which is herein described as Dugesia arabica Harrath and Sluys, sp. nov. This new species is mainly distinguishable from other Dugesia species that are distributed exclusively in the Mediterranean basin and in the eastern part of the Afrotropical region by the presence of the following features: well-developed and cone-shaped penis papilla, housing an ejaculatory duct that runs ventrally and has a subterminal and ventral opening; a considerably expanded and folded section of the bursal canal at the level of the oviducal openings; absence of a layer of longitudinal muscles on the copulatory bursa and the bursal canal. Specimens from two populations from Yemen were infested with a gregarine Protozoon.
In Europe, there are three aphid species of the genus Stomaphis Walker that feed on trees in the genus Acer and one that feeds on trees in the family Salicaceae. Although these species are considered morphologically distinct, their uncertain host specificity and variation in morphological features has led to misidentifications or questionable attribution of subspecies status. The aim of the present study was to clarify the identity of the Central European representatives on the basis of morphological and molecular analyses. Our study is based on 42 samples from Central Europe and Northern Italy. Mitochondrial markers COXI and COXII were used to determine the molecular identity of the specimens studied. Our molecular analyses revealed the existence of three clades (two on Acer, one on Salicaceae) as defined by both molecular markers. Morphological analysis showed a broad range of variability of the key morphological features in all three clades. Despite significant differences in the mean values of morphometric traits, their range of variability strongly overlapped. Samples from COXI clades on Acer showed two different patterns of geographical distribution. Host specificity is recognized only at the level of host plant genera (Acer) and family (Salicaceae), excluding the possibility of the existence of separate subspecies on single host plant species.
